Objective:The use of nano-gold rods has unique optical properties,stable and uniform dispersion,high surface activity,easy to combine with other atoms,can be sensitive and efficient detection of the characteristics of the sample to be prepared,the preparation of different diameter than the nano-gold bar solution to determine the preparation of surface plasmas The concentration and the maximum concentration of nano-gold rods.The detection of antigen and concentration of Trichinella spirala on the surface of Trichinella spiralis can be used to detect the serum antibody of Trichinella spiralis infection,and to provide the basic research for the rapid detection of Trichinosis.Method:1.Different concentrations,different amounts of CTAB,AA,Ag NO3 and different amounts of gold seed solution,the use of gold seed groth method to prepare different morphology of nano-gold rods.2.In the laboratory,the muscle tissue of Trichinella spiralis infected mice was detected by pepsin digestion method.The antigen of the Trichinella spiralis larvae was excised and the antigen was excreted.The antigen of the nano-gold rods solution was modified with different concentrations of antigen,and the antigen and excreted antigen.The surface modification of the wavelength changes to determine the surface modification of the appropriate concentration of the antigen.3.The serum antibody of different time,different degrees and different dilutions of Trichinella spiralis infected mice were detected by surface modification of Trichinella spiralis larvae with antigens.The results were compared with those of ELISA kits.Result:1.The gold nanoparticles with different morphologies were prepared by golden crystal seed growth method.The size of 0.1 M and 0.2 M CTAB could be used to prepare the nanometer gold bar solution with different particle size and stable structure.100 M concentration of ascorbic acid can be prepared uniform and stable nano gold rod solution.Different concentrations of silver nitrate can be different,the amount of cetyltrimethylammonium chloride,ascorbic acid,silver nitrate,gold seed seed liquid preparation can be obtained in different diameter ratio of gold gold bar solution.2.The crude antigen and excretory antigen of different concentrations of Trichinella spiralis were modified on the surface of nano-gold rods.The maximum peak of red peak was 8nm and 87 nm by UV spectrophotometer.3.The serum antibody was detected by detecting the excretory antigens of Trichinella spiralis on the surface of the trichinella spiralis.The detection of nano-gold bars was the first to appear on the 5th day.The positive results of the ELISA test showed the first 8 days.The maximum dilution of serum antibody was 1: 800,and the maximum concentration of positive antibody was 1: 700.The serum antibody of the mice with different infection degree was detected on the 5th day.The positive test was observed on the 5th day,and the lowest infection amount was 50.ELISA detection of the earliest positive is the first 8 days,the minimum infection is 100.Conclusion:1.The success of the use of seed growth by changing the growth solution in the protective agent,reducing agent,crystal seed concentration and the amount of different sizes of nano-gold rod2.Nano-gold bar surface modification preferred Trichinella spiralis larvae excretion of antigen,the best concentration of 20 ?g/m L.3.Nanoparticle labeling technique can detect sera antibodies from Trichinella corditis early.The first day after infection,the maximum dilution of serum antibody is 1: 800,and the minimum infection amount is 50,which is obviously superior to ELISA.
