Objective:By using flow cytometry check on glycoprotein in type 2 diabetes mellitus without complications group(DM group), simple type 2 diabetes mellitus complicated with diabetic peripheral neuropathy(DPN group) and normal subjects(NC group),it means that which can reflect the degree of platelet activation,the positive expression of platelet membrane GP:CD41,CD36, CD63, CD62 P, they can predict the risk of thrombus formation in microcirculation.The difference of the positive expression among three groups of four kinds of platelet membrane GP were compared, predict the platelet adhesion aggregation, the final formation of DPN,and provide more basis for prevention and treatment of DPN.Methods: Collected in our hospital during 2014-01-01 to 2014-12-30 health check-up crowd,50 healthy people were selected as NC group,50 cases in type 2 diabetes patients with no complications of DM group,the simple type 2 diabetes mellitus complicated with diabetic peripheral neuropathy patients group for DPN. Determination of the above three groups platelet membrane GP:CD41,D36,D63,D62 P with FCM positive expression level;at the same time,the ues of Toronto clinical scoring system(TCSS) score TCSS score was measured.determination of trace liver and kidney function, microalbuminuria,(m ALB),fibrinogen(FIB),platelet(PLT),platelet distributio n width(PDW), mean platelet volume(MPV), platelet-large cell ratio(P-LCR), fasting glucose(FPG), Hemoglobin A1C(Hb Alc); all diabetic patients underwent electrophysiological examination, namely sensory and motor nerve conduction of bilateral median nerve, ulnar nerve, tibial nerve, sural nerve,peroneal total nerve were determined by electromyography.In this study, the test data to SX ± that were normality and homogeneity of variance test,the differences between groups using SPSS version 13.0 software for the single factor analysis of variance,P<0.05 had significant difference.Results: 1 Platelet membrane GP in each group the positive expression rate of: 1.1 Positive expression rate of CD41 which was in NC group, DM group and DPN group was divided into: 97.44±3.40%, 97.16±2.45%, 97.17±3.16%, the positive expression rates of between the three groups had no statistical difference P> 0.05,F=0.137. 1.2 Positive expression rate of CD36 which was in NC group, DM group and DPN group was divided into:92.35± 4.52%, 94.08±3.87%, 95.43±1.73%, the three groups had statistical difference P<0.05,F=9.308. There was significant difference between NC group and DM group,NC group and DPN group, P<0.05; there was no significant difference between DM group and DPN group,P>0.05. 1.3 Positive expression rate of CD63 was divided into: 2.61±0.90%, 4.24± 2.91%, 7.65±3.79%, the three groups had statistical difference P<0.05, F=42.073. There were significant differences between NC group and DM group, NC group and DPN group, DM group and DPN group, P< 0.05. 1.4 Positive expression rate of CD62 P was divided into:1.70±0.84%, 3.31± 2.27%, 6.92±3.60%, the three groups had statistical difference P<0.05, F=57.107. There were significant differences between NC group and DM group, NC group and DPN group, DM group and DPN group, P< 0.05.In summary 1) The positive expression of CD36,CD63,CD62 P rate of T2 DM group, DPN group were higher than those in NC group. 2) The positive expression of CD36, CD63, CD62 P the rate difference between the three groups, DM group, DPN group were higher than those in group NC.3) CD62 P and CD63 the positive expression rate of there were differences between T2 DM and DPN group,DPN group higher than that of DM group. 4)The positive rate of CD36 was no difference in DM group with DPN group. 2 NC group, DM group and DPN platelets between each index respectively: 2.1 PLT(* 109/L): 233.78±51.06,227.84±49.17,235.60±62.74, there was no significant difference between these three groups,P>0.05,F=0.276. 2.2 PDW: 10.97±1.23,10.90±0.93,10.66±0.99,there was no significant difference between these three groups,P>0.05,F=1.202.There was no significant difference between NC group and DM,NC group and DPN group, DM group and DPN group, P>0.05. 2.3 MPV(f L):12.25±2.10,12.97±1.66,12.35±1.83, there was no significant difference between these three groups,P>0.05,F=2.191. There was no significant difference between NC group and DM,NC group and DPN group, DM group and DPN group, P>0.05. 2.4 P-LCR(%):28.71±7.00,31.48±6.81,28.44±7.28, there was no significant difference between these three groups,P>0.05,F=2.855.There was no significant difference between NC group and DM group, NC group and DPN group, P>0.05; there was significant difference between DM group and DPN group(P<0.05). 3 FIB(g/L) in the NC group,DM group,DPN group index was divided into: 2.88±0.39,3.05±0.66,3.16±0.87, there was no significant difference between these three groups,P>0.05,F=2.146.There was no significant difference between NC group and T2 DM group,P>0.05;there was significant difference between NC group and DPN group,P<0.05; there was no significant difference between DM group and DPN group,P>0.05. 4 Correlation analysis among the positive expression rate of platelet membrane glycoprotein and BMI、Crouse、HBA1C、SBP、DBP、TC、TG、HDL-C、LDL-C、PLT、PDW、MPV、P-LCR、FIB.The results:CD62P is directly proportional to BMI(r=0.341,P<0.05),and notable directly proportional to PDW(r=0.381,P<0.05), directly proportional to MPV(r=0.318,P<0.05), directly proportional to P-LCR(r=0.317,P<0.05), notable directly proportional to CD63(r=0.474,P<0.05).Conclusion:1 DM group and DPN group patients platelet membrane GP:CD36,CD63, positive expression rate of CD62 P increased in the NC group,it suggested that platelet activation in patients with type 2 diabetes mellitus and diabetic peripheral neuropathy,it increased risk of platelet adhesion, aggregation and thrombosis.2 Platelet membrane GPCD41 could not predict the platelet activation, thrombosis risk and risk of DPN; CD36 was also can not be predicted to occur as the risk of diabetic peripheral neuropathy in patients with type 2 diabetes mellitus; CD63,CD62 P prompt that platelet membrane GP activation that DPN group was enhanced in the DM group, platelet adhesion, aggregation, thrombosis risk was greatly increased.3 The traditional use of the observation of platelet function index:PLT, PDW, MPV, P-LCR, FIB were not different from the NC group,DM group,DPN group, that suggested that these indicators in the DPN group no excessive expression,it could not well the DPN group the activation of platelet adhesion, aggregation and thrombosis risk,it could not reflect the activation of DPN.
