Objective: The purpose of this study is to investigate the correlation between the change of 3-nitrotyrosine (3-NT) and Glutathione-peroxidase (GSH-PX) for diabetic peripheral neuropathy in patients with type 2 diabetes mellitus and its development. To investigate the possibility mechanism of 3-NT and GSH-PX in diabetic peripheral neuropathy in patients with type 2 diabetes mellitus, to provide new effective idea for diabetic peripheral neuropathy in patients with type 2 diabetes mellitus.Methods: Selected the patients and healthy volunteers in our hospital which are from the department of endocrinology and health-examination, research objects is divided into two groups:simple control (NO group 20 cases NC, and type 2 diabetes mellitus (T2DM) group 64 cases, among the total of the 64 cases were NDPN group 32 cases and DPN group 32 cases. To collect all research objects fasting elbow vein blood. Adopt double antibody sandwich enzyme linked immunosorbent assay (ELISA) measure blood plasma 3-NT, blood plasma GSH-PX, at the same time collected the fasting blood glucose (FBG),blood lipid, hemoglobin A1 of glycosylated (HbAic), fasting insulin (FINS),fasting C-peptide (FC-P) and other biochemical index sign. The same period records the sex,age, weight, height,body mass index (BMI),waist-hip ratio (WHR),course of diabetes,blood pressure and etc. The vibration threshold value of both lower extremities and nerves electroneuromyography were used for screening all cases. Carry on the data processing with the SPSS 17.0 statistics software, data express with meanĀ±tandard, multigroups comparison by analysis of variance (ANOVA), two group comparison by t-test, multiple factors use gradually linear regression analysis, use the multiple Logistic regression analysis the relationship among every index.Results:(1) Blood plasma 3-NT of DPN group were obviously higher than group NDPN and NC group, they were significantly difference (P<0.05); then blood plasma 3-NT have no difference between NC group and NDPN group (P<0.05);(2) Blood plasma GSH-PX of DPN group were obviously lower than group NDPN and NC group, they were significantly difference (P<0.05); then GSH-PX of NDPN group were lower than NC group, they were significantly difference (P<0.05);(3) DNP in patients with type 2 diabetes were correlate to the 3-NT,GSH-PX,Course of diabetes,FBG, age(P<0.01);(4) Through the multiple Logistic regression analysis we found that the blood plasma 3-NT,Course of diabetes,age were risk factors to DPN of type 2 diabetes mellitus; And the blood plasma of GSH-PX were protectant factors to DPN of type 2 diabetes mellitus.Conclusion: Blood plasma 3-NT of DPN group were obviously higher than group NDPN and NC group, then blood plasma GSH-PX of DPN group were obviously lower than group NDPN and NC group; displayed that 3-NT and GSH-PX have closed correlation in occurrence and development of diabetic peripheral neuropathy in patients with type 2 diabetes mellitus.
