Objective Using RNA interference, RNA interference, RNAi technology silence after the Cat S gene in liver cancer cells, then observe the changes of its sensitivity to Adriamycin.Methods 1.design a pair for Cat S gene of siRNA, transfection human liver cancer cells MHCC97-h, targeted Cat S gene silence as experimental group, at the same time set up empty carrier of the negative control group and blank group (not transfection), western blot detection CatS expression in silence after three groups.2.Give different concentrations of more soft than star in more than one time period to cultivate the experimental group and negative control cells, determined by MTT selected two groups of cells inhibition rate difference biggest pair for next experiment.3.Western bolt testing this group of apoptosis protein expression of Caspase 3 levels.Results 1.RNAi technology silent Cat S gene in human liver cancer cells MHCC97-H after the Cat S expression in the experimental group, compared with the control group and the blank group, p values were 0.000 (p<0.05), statistically significant, while the control group compared with the blank group, no difference (p> 0.05).2.when the concentration of Adriamycin reached 32 ug/ml intervention after 24 hours, the experimental group and the negative control group cell inhibition rate difference biggest p1.58 E-5p<0.01), and as Adriamycin concentration and intervention time increases, the two groups of cells inhibition rate on the rise, when the concentration of 64 ug/ml intervention after 36 hours, two groups of cell apoptosis.3, Adriamycin concentration is 32 ug/ml intervention group and control group after 24 hours, compare the level of apoptosis protein Caspase 3.p0.000 experimental group is significantly higher than the negative control group (p<0.05), the difference was statistically significant.Conclusions Silent Cat S gene expression in human liver cancer cells can enhance sensitivity to Adriamycin.
