Objective:To prepare and evaluate the alginate hydrogel bilayer films containing Aloe vera for clinical applications of natural hydrogel material.Methods:The hydrogel bilayer films were prepared by chemical crosslinking alginate and Aloe vera gel in different proportions. Chemical composition, transparency, mechanical properties were studied by ATR-FTIR, UV, Universal testing machine. The swelling behavior, degradation behavior of hydrogel was studied by observing its weight change. Its antibacterial activity was evaluated by inhibition zone experiments. The adhesion and proliferation of L929 cells were observed by MTT and fluorescent staining. In addition, skin irritation and sensitization were also evaluated by SD rats.Results and Conclusion:1) ATR-FTIR spectra showed characteristic infrared absorption peaks of Aloe vera, indicating alginate hydrogel bilayer films containing Aloe vera were prepared successfully. The films had smooth surface, high transparency and good mechanical properties in contrast to control group. Meanwhile, this hydrogel had pH sensitive property, and it showed the high swelling ratio (1223%) at pH= 7.4, its degradation rate could be improved by the introduction of Aloe vera.2) Antibacterial test suggested that the hydrogel films containing Aloe vera had bacterial inhibitory abilities to E.coli and S.aureus, as the films appeared antibacterial activity while the Aloe vera content was more than 15%.3) Cytocompatibility experiment showed that the hydrogel films could promoted L929 cells proliferation and adhesion, and the relative growth rate attained the highest value of 163% (p< 0.05), so the cytotoxicity can be classified to zero degrade.4) The result of stimulation experiment and allergy experiment indicated that this material has weak allergic responses and no skin-irritation.
