The Citrus genus is one of the most widely cultivated horticultural crops in the world. China is the principal origin centre of Citrus. Some important wild citrus species originated from this country where gathered a large number of citrus processing enterprises. Citrus peels contain a variety of bioactive substances beneficial to our health such as flavonoids, and phenolic acids. It has been wildly reported that the citrus flavonoids have antioxidant activity and anti-inflammatory activity. Citrus bioactivity has become a worldwide hot spot research.In present study we choose 7 genotypes of Chinese citrus which with high levels of flavonoids in their peels. their name as follow, Wulongsuanju, Nieduyeju, Limeng, Taiwanpenggan, Guangxishatianou, Edanhongju and Qingjainjucheng. We compare the flavonoids content and antioxidant activity and anti-inflammatory activity of different polar fraction of ethanol extraction of 7 genotypes of citrus peels. The purpose is to find out the preferable antioxidant and efficiency anti-inflammatory genotype. In present study, we evaluated the antioxidant and anti-inflammatory activity of different polar fractions from primary ethanol extract of citrus. We study the flavonoids of activity of petroleum ether fraction (PEF), ethyl acetate fraction (EAF), n-butanol fraction (NF), and aqueous fraction (AF) of fractions were evaluated by HPLC. Antioxidant activity was estimated using 2,2-diphenyl-l-picrylhydrazyl (DPPH) and 2,20-azinobis (3-ethylbenzothiazoline-6-sulfonate) (ABTS) free radical scavenging assays and oxygen radical absorbance capacity assay (ORAC). The anti-inflammatory activity was assessed using RAW264.7 cells by NO assay and enzyme linked immunosorbent assay (ELISA).The mean results are as follows:1. The calculations of total flavonoids indicate that the EAF has the highest flavonoid content, followed by NF and PEF. The highest flavonoids content of EAF was found in Nieduyeju, followed by Wulongsuanju, Taiwanpenggan, Guangxishatianyou, Qingjianjucheng, Limeng, and Edanhongju. And each EAF was form 73.29%,65.38%,67.35%,71.04%, 57.34%,74.84%,53.5% of the total flavonoids, separately. The flavonoids contents of different EAF are variety, the Nobiletin are the highest contents of EAF of Nieduyeju, Taiwanpenggan and Wulongsuanju, it was form 44.90%,76.26% and 49.02% of the total EAF flavonoids, separately.2. The antioxidants activities of different polar fractions from primary ethanol extract of citrus are positively correlated with flavonoids contents. the ethyl acetate extract of the strongest antioxidant capacity, n-butanol, followed by petroleum ether and the aqueous phase is weak parts of antioxidant capacity.he rank of EAF parts of ACP as follows Nieduyeju> Wulongsuanju> Taiwanpenggan> Qingjianjucheng> Guangxishantianyou> Limeng> Edanhongju. Citrus peel ethanol extract antioxidant capacity of different polar fractions and flavonoid content was significantly positively correlated.3. The trends of anti-inflammatory activities of different polar fractions from primary ethanol extract of citrus are positively correlated with flavonoids contents. EAF treatment noticeable inhibited the LPS-stimulated NO production in a dose-dependent manner. EAF of Nieduyeju, Wulonguanju and Taiwangpenggan appeared to be the most efficient and induced significant decrease of NO release (P< 0.05) comparing to control. ELISA test showed that EAF of Nieduyeju, Taiwangpenggan and Wulonguanju could significantly inhibited secretion of the cytokine TNF-a, IL-6, IL-10. Nobiletin may plays an important role in the anti-inflammatory activity.
