Objective:To detect the expression of YB-1, P53 in chronic cervicitis, CINI, CINII-III and cervical squamous carcinoma tissues, investigate the relationship between the expression of YB-1, P53 and the growth and metastasis of cervical squamous carcinoma, and to analyze their interaction.Methods:Collected 15 cases of chronic cervicitis, 15 cases of CINI, 30 cases of CINII-III and 45 cases of cervical squamous carcinoma, Immunohistochemistry(IHC) Envision was used to detect the expression of YB-1 and P53. All data use SPSS17.0 analysis: Chi-square test or Fisher’s exact test were used to identify the difference among groups for frequency data, Wilcoxon singned-rank test was used to identify the difference among groups for Rank data, The relationship was analyze by Spearman rank correlation.Results:1. YB-1 was mainly located in the nucleus in squamous cell, sometimes in the cytoplasm, P53 protein is locted in the nucleus in squamous cell. 2. The YB-1 protein did not expression in chronic cervicitis. In CINI, CINII-III and cervical squamous carcinoma, the positive expression rates were 6.67%, 20.00%, 62.20%. 3.The four groups compared, the expression of YB-1 was satistically significant(χ2=33.45,P=0.000). The chronic cervicitis compared with the cervical squamous carcinoma, the difference was statistically significant(χ2=16.71, P=0.000); the CINI compared with the cervical squamous carcinoma, the difference was statistically significant(χ2=14.49, P=0.001); the CINII-III compared with the cervical squamous carcinoma, the difference was statistically significant(χ2=17.49, P=0.000). From spearman rank correlation analysis: the expression of YB-1 was positively correlated with degree of cervical lesions(rs=0.559,P=0.000). 4. In the P53 protein, the positive expression rates in chronic cervicitis, CINI, CINII-III, cervical squamous carcinoma were 6.67%,13.33%,53.33%,66.67%. 5. The four groups compared, the expression of P53 was satistically significant(χ2=30.41,P=0.000). The chronic cervicitis compared with the CINII-III, the difference was statistically significant(χ2=8.52,P=0.018); the chronic cervicitis compared with the cervical squamous carcinoma, the difference was statistically significant(χ2=16.31,P=0.000); the CINI compared with the CINII-III, the difference was statistically significant(χ2=6.61,P=0.054); the CINI compared with the cervical squamous carcinoma, the difference was statistically significant(χ2=14.90, P=0.001); the CINII-III compared with the cervical squamous carcinoma, the difference was statistically significant(χ2=7.25, P=0.062). From spearman rank correlation analysis: the expression of P53 was positively correlated with degree of cervical lesions(rs=0.502,P=0.000). 6.In the cervical squamous carcinoma group, there were no correlation between the expression of YB-1 and the age, parametric infiltration, lymph node metastasis, tumor size and the degree of differentiation in cervical squamous carcinoma, but they are associated with clinical stage and depth of stromal invasion. The expression of YB-1 in clinical stageII was significantly higher than clinical stageI(Z=-1.729,P=0.084), the deep muscle layer and the whole infiltration was significantly higher than shallow muscular infiltration(Z=-1.820,P=0.069). 7. In the cervical squamous carcinoma group, there were no correlation between the expression of P53 and the age, depth of stromal invasion, parametric infiltration, lymph node metastasis, tumor size and the degree of differentiation in cervical cancer, but they are associated with clinical stage. The expression of P53 in clinical stageII was significantly higher than clinical stageI(Z=-1.83,P=0.067). 8. There was no correlation between the expression of YB-1 and P53 in cervical squamous cell carcinomas(rs=-0.142,P=0.351).Conclusion:1. In differert cervical tissues, The expression of YB-1 and P53 showed increasing trend, they are associated with the development of cervical squamous carcinoma. 2. The expression of YB-1 was related with different clinical stage and different depth of stromal invasion, but there were no correlation with the age of the patients, vascular invasion, lymph node metastasis, tumor size and the degree of differentiation in cervical squamous carcinoma. 3. The expression of P53 was related with different clinical stage, but there were no correlation with different depth of stromal invasion, the age of the patients, vascular invasion, lymph node metastasis, tumor size and the degree of differentiation in cervical squamous carcinoma. 4. The expression was no correlation between the YB-1 and P53.
