Objective:To detection the expression of cyclin-dependent kinase 2(cyclin-dependent kinase 2, CDK2) and cell cycle protein E(Cyclin E) in meningioma through the immunohistochemistry method, explore the relationship between the expression of CDK2 and Cyclin E in meningioma and the clinical performance,pathology of meningioma.Method : To detect expression of CDK2 and Cyclin E in normal brain tissue and meningioma tissues through the immunohistochemical method(SP method),analysis of the experimental results combined with clinical data.Result:(1)4 cases of meningeal tissue with CDK2 positive expression out of 20 cases(positive rate 20%),45 cases of meningiomas with CDK2 positive expression out of 64 cases(positive rate 70.31%),in which weakly positive, moderate positive and strong positive expression of CDK2 were26 cases,12 cases, 7 cases. The difference between the expression of CDK2 in meningeal tissue and meningioma tissues was significant(Z=-4.033,P=0.000<0.05).(2)4 cases of normal arachnoid tissue with CDK2 positive expression out of 20 cases(positive rate 20%),38 cases with positive expression of CDK2 in meningiomas of grade I, in which weakly positive, moderate positive and strong positive expression were 25 cases, 10 cases and 3 cases。7 cases with positive expression of CDK2 in meningiomas of grade Ⅱ, in which weakly positive, moderate positive and strong positive expression were 1cases, 2 cases and 4cases. The difference between the expression of CDK2 in grade I meningiomas and meningeal tissue was significant(Z=-3.678,P=0.000<0.05);The difference between the expression of CDK2 in gradeⅡ meningioma and meningeal tissue was significant Z=-4.252,P=0.000<0.05);The difference between the expression of CDK2 in grade I meningiomas and gradeⅡ meningioma was significant(Z=-3.45,P=0.001<0.05)。(3)Pathological grading was positively correlated with CDK2 expression in meningioma(rs=0.434,P=0.000<0.05);(4)In the 64 cases of meningioma patients,25 cases are male, in which negative,weakly positive, moderate positive and strong positive expression of CDK2 were 7 cases,10 cases, 10 cases and 3 cases;39 cases are female,in which negative,weakly positive, moderate positive and strong positive expression of CDK2 were 12 cases,16 cases, 7 cases and 4 cases. The expression of CDK2 in meningioma was not raleted with patient’s gender(Z=-0.334,P=0.739>0.05)。(5)There were 64 cases of meningioma in the experimental group,in which,sagittal sinus meningioma,convex meningioma,sphenoid ridge meningioma,falx meningioma,saddle nodule meningioma,olfactory sulcus meningioma,trigone pontocerebellar pontocerebellare trigonum meningioma,cerebellar meningioma are 16 cases,12 cases,9 cases,8 cases,6 cases,6 cases,4 cases and 3cases.The expression of CDK2 in meningioma was not raleted with the site of meningioma(Z=0.511,P=0.999>0.05).(6) 7 cases that diameter less than or equal to 3cm in the 64 cases of meningioma, in which negative,weakly positive, moderate positive andstrong positive expression of CDK2 were 2 cases,4 cases, 1cases and 0 cases;35 cases that diameter between 3cm and 5cm in the 64 cases of meningioma, in which negative,weakly positive, moderate positive and strong positive expression of CDK2 were 14 cases,11 cases, 5cases and 5 cases;22 cases that diameter greater than or equal to 5cm in the 64 cases of meningioma, in which negative,weakly positive, moderate positive and strong positive expression of CDK2 were 3 cases,11 cases, 6cases and 2 cases. The expression of CDK2 in meningioma was not raleted with the size of meningioma(c2=2.423,P=0.298>0.05).(7) 6 cases of normal meningeal tissue with Cyclin E positive expression out of 20 cases(positive rate 30%),49 cases of meningiomas with Cyclin E positive expression out of 64 cases(positiverate76.56%),in which,weakly positive, moderate positive and strong positive expression of Cyclin E were26 cases,12 cases, 7cases. The difference between the expression of Cyclin E in meningeal tissue and meningioma tissues was significant(Z=-3.963,P=0.000<0.05).(8) 6 cases of normal meningeal tissue with Cyclin E positive expression out of 20 cases(positive rate 30%),42 cases with positive expression of Cyclin E in meningiomas of grade I, in which weakly positive, moderate positive and strong positive expression were 22 cases, 16 cases and 4 cases.7 cases with positive expression of Cyclin E in meningiomas of grade Ⅱ, in which weakly positive, moderate positive and strong positive expression were 1cases, 1 cases and 5 cases。The difference between the expression of Cyclin E in grade I meningiomas and meningeal tissue was significant(Z=-3.327,P=0.001<0.05);The difference between the expression of Cyclin E in grade Ⅱ meningiomas and meningeal tissue was significant(Z=-3.906,P=0.000<0.05); The difference between the expression of Cyclin E in grade I meningiomas and gradeⅡ meningioma was significant(Z=-3.29,P=0.001<0.05).(9) Pathological grading was positively correlated with the expression intensity of Cyclin E in meningioma(rs=0.414,P=0.001<0.05);(10)In the 64 cases of meningioma patients,25 cases are male, in which negative,weakly positive, moderate positive and strong positive expression of Cyclin E were 6 cases,9 cases, 7 cases and 3 cases;39 cases are female,in which negative,weakly positive, moderate positive and strong positive expression of Cyclin E were 9 cases, 14 cases, 10 cases and 6 cases. The expression of Cyclin E in meningioma was not raleted with patient’s gender(Z=-0.179,P=0.858>0.05).(11) There were 64 cases of meningioma in the experimental group,in which,sagittal sinus meningioma,convex meningioma,sphenoid ridge meningioma,falx meningioma,saddle nodule meningioma,olfactory sulcus meningioma,trigone pontocerebellar pontocerebellare trigonum meningioma,cerebellar meningioma are 16 cases,12 cases,9 cases,8 cases,6 cases,6 cases,4 cases and 3cases.The expression of Cyclin E meningioma was not raleted with the site of meningioma(Z=2.661,P=0.915>0.05).(12) 7 cases that diameter less than or equal to 3cm in the 64 cases of meningioma, in which negative,weakly positive, moderate positive and strong positive expression of Cyclin E were 1 cases,4 cases, 2cases and 0cases;35 cases that diameter between 3cm and 5cm in the 64 cases of meningioma, in which negative,weakly positive, moderate positive and strong positive expression of Cyclin E were 10 cases,13 cases, 6 cases and 6 cases;22 cases that average diameter than or equal to 5cm in the 64 cases of meningioma, in which negative,weakly positive, moderate positive and strong positive expression of Cyclin E were 4 cases,6 cases, 9 cases and 3 cases. The Expression of Cyclin E in meningioma was not raleted with the size of meningioma(c2=1.469,P=0.480>0.05).(13)Negative,weakly positive, moderate positive and strong positive expression of CDK2 and Cyclin E were 6 cases,12 cases, 4 cases and 3 cases, the relationship of the expression of CDK2 and Cyclin E in meningioma is positive(rs=0.323,P=0.009<0.05). Conclusion:(1)The differences of the expression of CDK2 and Cyclin E between normal arachnoid tissue and meningioma were significantly,which concluded that CDK2 and Cyclin E may be involved in the occurrence, development of meningioma.(2)The expression of CDK2 and Cyclin E were not raleted with patient’s gender and the site,size of meningioma;(3)The expression of CDK2 and Cyclin E were associated with the pathological grading of meningioma.( 4) The relationship of the expression of CDK2 and Cyclin E in meningioma is positive,there may be a positive interaction. CDK2 and Cyclin E have a close relationship with the clinical performance, pathology of meningiomas.A further study of the expression CDK2 and Cyclin E and its mechanism in meningiomas may find out a new therapy for the treatment of meningiomas.
