| Background Polycystic ovary syndrome (PCOS) is an exceptionally common disorder of premenopausal women characterized by hyperandrogenism, increased luteinizing hormone/follicle stimulating hormone (LH/FSH) ratio and excessive insulin. Its pathogenesis is believed to be multifactorial, and abnormal gene regulation could be one contributing factor. The polymorphism in the FSHβ promoter (-211G→T, rs10835638) appears to be associated with elevate FSH in females, FSH is a gonadotropins, among women, its main physiological function are:(1) directly promoting befor sinus follicle and follicular granulosa cell proliferation and differentiation, the follicle growth and development; (2) the activation of granular cell aromatase, plays an important role for testosterone into estrogen; (3) to adjust the choice of dominant follicle and the Atresia degradation of minor follicle; (4) in the late follicular phase,with estrogen, inducing granulosa cells generating LH receptor, preparing for ovulation and su-hua huang.So far,no data on consequence of FSHβ promoter -211G→T is the possible cause of PCOS.Objective Whether FSHβ-211G→T mutation influence PCOS susceptibility and phenotypes or not.Methods A total of 254 women (100 patients with PCOS and 154 healthy controls) were recruited at Department of Gynecology, No.174 hospital of PLA, Xiamen between March 2013 and August 2013 in the study. We recorded a standardized evaluation that included clinical data on age, body height, weight, BMI, waist and hip,etc.The peripheral blood were obtained on the third day of the menstrual cycle,which was used to determine the reproductive hormone, including FSHã€LHã€E2ã€Pã€TTã€PRL.Fasting blood glucose and fasting serum lipid were determined in peripheral blood between 06:30 and 07:00 a.m. after an overnight fasting. The genotypes of FSHβ-211G→T gene were determined by PCR and DNA sequencing.Results 1.Clinical variables of controls and PCOS women were summarized.The LH, LH/FSH, Fasting insulin and triglycerides levels were higher in PCOS patients compared with those of controls, as same as Systolic blood pressure, Waist, Hirsutism score and Acne score. However, the SHBG and HDL levels were significantly lower in PCOS patients compared with those of controls(P<0.05).2.The FSHβ-211G→T genotype frequency in PCOS was significantly higher than that in controls (P=0.046).3. In terms of FSHβ-211G→T, the LH/FSH were significantly different among GG and GT genotypes in PCOS patients and controls, exhibiting higher LH/FSH in the GT genotype(p=<0.05). FSH levels were significantly lower in the GT genotype in PCOS patients (p=<0.05), whereas LH/FSH and cholesterol levels were significantly higher in the GT genotype in controls(p=<0.05).4. FSHP-211G→T genotype frequency in the overall study population and PCOS or control,having no effect on BMI, T, hairy and insulin resistance (P>0.05).Conclusions Results suggested FSHβ-211G→T mutation might influence PCOS susceptibility. Also we found mutation genotype GT of FSHβ-211G→T may have effete on women’s reproductive hormones,such as FSH, LH, etc. in order to further the cause of PCOS,our research provides a new basis. |
