Effect Of Jiawei Qigong Pill On Follicle Stimulating Hormone And Luteinizing Hormone In Obese Polycystic Ovary Syndrome Model Mice

Objective: This study focus on the development of the obese polycystic ovarian syndrome(PCOS)model mice,which are treat in jiawei qigong pill to observe the effects of this pill on body weight,luteinizing hormone(LH),follicle stimulating hormone(FSH)and ovarian tissue morphology.Methods: In this study,56 SPF 23 day old female mice were randomly divided into two groups,including 13 normal control group and 43 model group.Module to dehydroepiandrosterone(DHEA)joint chain urea with streptozotocin(STZ)and high fat and sugar feed obese PCOS model mice.After successful modeling,mice in the modeling group were randomly divided into low-dose group,medium-dose group,high-dose group and PCOS control group.PCOS control group and normal group were given distilled water once a day,and each treatment group was given the corresponding dose of jiawei qigong pill once a day,and each group was given continuous gavage for 15 days.After the intervention of jiawei qigang pill,serum and ovarian tissues of each group were collected,radioimmunoassay(RIA)was used to detect LH and FSH levels in serum of mice,and the histological changes of ovary were observed by HE staining.Results:1.Effect of Jiawei Qigong Pill on the Body Weight of Obese PCOS Model MiceAfter treatment,the body weight of PCOS control group is(34.40±1.16)g,low dose group is(33.83±0.49)g,medium dose group is(34.40±1.57)g,high dose group is(31.85±1.17)g,and normal group is(24.95±1.71)g.After statistical analysis,the body weight of mice in the low-dose group,the medium-dose group and the high-dose group increases compares with the normal group(P < 0.05).There is no significant difference in body weight between the low-dose group and the medium-dose group and the PCOS control group(P > 0.05),while the body weight of the high-dose group is significantly reducing compares with the PCOS control group(P < 0.05).2.Effect of Jiawei Qigong Pill on Serum LH and FSH in Obese PCOS Model MiceAfter treatment,LH of PCOS control group is(329.57±9.58)mIU/ml and FSH is(1.87±0.29)mIU/ml.In the low-dose group,LH is(72.60±0.89)mIU/ml and FSH is(3.95±0.69)mIU/ml respectively.In the medium dose group,LH is(68.13±3.52)mIU/ml,and FSH is(2.87±0.28)mIU/ml.In the high-dose group,LH is(79.85±13.31)mIU/ml,and FSH is(3.24±0.50)mIU/ml.LH in the normal group is(63.77±2.67)mIU/ml,and FSH is(2.53±0.17)mIU/ml.After statistical analysis,the serum LH and FSH of the low-dose group,the medium-dose group and the high-dose group are significantly reducing(P<0.05)and the serum FSH is significantly increasing(P<0.05),which are all statistically significant.3.Effect of Jiawei Qigong Pill on Ovarian Histomorphology of Obese PCOS Model MiceUnder the microscope,follicles and luteal bodies at different developmental stages are observing in the normal group,and the granular layer cells are closely arranging.The ovarian tissue of PCOS control group show more follicular dilatation and atresia follicles under microscope,with disordered arrangement of granulosa cells and decreased number of corpus luteum.After treatment with jiawei qigong pill,the number of original follicles under microscope in the ovarian tissue of mice in the low-dose group decrease,the number of corpus luteum increased compares with the PCOS control group,and the arrangement of granulosa cells is more disordered than that in the normal group.The number of follicles in the medium-dose group is decreasing compares with that in the PCOS control group.The mice in the high-dose group has smaller corpus luteum than those in the normal group.Conclusion: Jiawei Qigong Pill can improve serum LH and FSH levels and ovarian tissue morphology in obese PCOS mice.
