Objective:To study growth factors expression of alkaline fibroblast growth factor (bFGF) and vascular endothelial growth factor (VEGF) of high-energy narrow spectrum red light on chronic painless wound model rats.Methods:45 rats were randomly divided into trauma model without exposure group, the 640 nm red short-term exposure group (30 min) and long-term exposure group (60 min),15 in each group. The three groups with 10% chloral hydrate intraperitoneal injection of 300 mg/kg anesthesia, formed in the spine using circular drill 4 to 10 mm diameter circular wound. The output optical power semiconductor laser were set in 640 nm. This experiment irradiation time were set every time 30 min and 60 min, respectively,1 times a day and a total of 14 times. Irradiation rats was every time according to position and the relative position of the light beam are remain the same. Every 2 wound were photographed using computer Image analysis software to measure the wound area by Image-ProPlus 6.0 software and a total of eight times were calculated the rate of trauma surface shrinkage. The wound tissue were cut and soaked in 25% formalin solution to finalize for 24 hours, made paraffin section, stained conventional HE, and then comparatively observed in the treatment of the 3 day,7 days, and 14 days. bFGF and VEGF of paraffin section were tested by immunohistochemical staining detection, collected immunohistochemical slicing digital photos, and calculated expression area. The healing rate of wound area were analyzed and calculated with image processing software Motic pro 5.0.Results:The 30 min exposure group,60 min exposure group compared with no radiation group showed statistically significant difference in the wound healing rate(P < 0.05)by repeated variance analysis. The 30 min exposure group,60 min exposure group compared with no radiation group, the area of wound healing rate showed statistically significant difference(P< 0.05). The 640 nm red long exposure group and short-time exposure group compared with model group, the bFGF expression quantity showed significant differences(P< 0.01), but the 640 nm red long exposure group and short-time exposure group had no significant difference (P> 0.05). The 640 nm red long exposure group and short-time exposure compared with model group, the expression quantity of VEGF showed significant differences(P< 0.01), but the 640 nm red long exposure group compared with short-term exposure group had no significant difference(P> 0.05).Conclusion:high-energy narrow spectrum red light treatment can obviously promote wound healing, improve the healing rate, improve the healing quality. The mechanism of promoting wound healing of Shengjibaiyu ointment may be associated with the modulation of cutaneous wound granulation tissue in bFGF, VEGF expression.
