AimIL- 10 is the important anti-inflammatory factor, play the role of anti-inflammatory and regulation through combine with ligand IL – 10 R. Recent research has found that in the patients with HBV infection, expression level of IL-10 R increased markedly.Hence, this experiment examine whether IL-10 R polymorphisms were associated with susceptibility to chronic HBV infection in a Chinese Han population through the study with two tag SNP: rs2228055、rs2256111 of IL-10 R.Methods Collecting 501 cases of Peripheral blood specimens with patients with chronic HBV infection of Chinese han population as case group, 301 cases of peripheral blood specimens with self-limiting HBV-infected individuals as control group. Two groups of patients’ age, gender and regional sources are match each other. The standard set of Chronic HBV infection refered to the diagnosis of < Chronic hepatitis B prevention and treatment guidelines > joint revised by the Chinese medical association liver disease, and infection of neurology in 2010. The standard set of self-limiting HBV-infected individuals were no history of hepatitis b vaccination,, Hbs Ag negative, HBs Ab、HBc Ab positive,HBV-DNA negative,ALT and AST all within the normal range,and all positive after two reexamination. Collected Peripheral Venous Blood of subject to extracte of genomic DNA,find two tag SNP:rs2228055、rs2256111 in the Hapmap database of IL-10 R gene.Then detectived two tag SNP genotypes with Snapshot..Finally,the frequencies of alleles and genotypes between two groups were analyzed by χ2 analysis.Results1. All genotype and allele distributions in IL – 10 R genes rs2228055、rs2256111 were in Hardy–Weinberg equilibrium. Two groups of patients’ age, gender and regional sources are match each other. 2. IL – 10 R gene tag SNP rs2228055 site genotype AA、GA、GG in the frequency distribution of C-HBV were 49.9%(250/501)、42.9%(215/501)、7.2%(36/501) respectively.In SL-HBV were 51.5%(155/301)、35.5%(107/301)、13.0%(39/301)。Overall haplotype P values were: AA,P=0.662, OR[95% confidence interval(CI)]=0.94(0.71-1.24); GA, P=0.039, OR[95% confidence interval(CI)]=1.36(1.02-1.83); GG, P=0.007, OR[95% confidence interval(CI)]=0.52(0.32-1.38). The frequency of GG genotype(7.2%) was significantly decreased in patients with C-HBV as compared to SL-HBV patients(13.0%). There is statistically significant difference between cases and controls(P<0.05); GG genotype in the risk of chronic HBV infection is 0.52 times of AG + AA genotype(OR=0.52,95%CI:0.32-0.84). tag SNP rs2256111 site genotype AA、GA、GG in the frequency distribution of C-HBV were10.6%( 53/501),46.1%(231/501),43.3%(217/501). In SL-HBV were 15.6%(47/301), 37.9%(114/301), 46.5%(140/301). Overall haplotype P values were: AA,P=0.037,OR[95% confidence interval(CI)]=0.64(0.42-0.98);GA, P=0.023, OR[95% confidence interval(CI)]=1.40(1.05-1.88);GG,P=0.378,OR[95% confidence interval(CI)]=0.88(0.66-1.17)。The GA genotype in patients with chronic HBV infection has a tendency to rise,but there is no statistically significant difference between cases and controls through Bonferroni correction. 3. Haplotype analysis and comparison, two polymorphic loci build the haploid through SHEsis,there is no statistically significant difference between cases and controls. Pc values were: AA,PC=0.387,OR(95%CI)=1.24(0.76-2.02);AG,PC=0.781, OR(95%CI)=1.04(0.84-1.29); GA, PC=0.387, OR(95%CI)=0.92(0.73-1.14)。ConclusionIn this study, the results suggested that the rs2228055 GG genotype of IL-10 R was associated with the susceptibility to chronic HBV infection in the Chinese Han population. Genetic polymorphism of IL-10 R may participate in the regulation and control of Chronic HBV infection。...
