AIM To investigate the association between the three IRF-3gene tagging singlenueleotide polymorphisms sites(tagSNPs)(rs10415576,rs2304204and rs2304206) andgenetic susceptibility to hepatitis B virus (HBV) infection in Chinese patients.Methods We collect985case of chronic HBV infection patients as cases and294caseacute self-limiting of HBV infection recovery period individuals as controls. All sampleswere collected from December2009to September2010; Based on the case-control study,985cases of chronic HBV infection as cases and294acute self-limiting HBV infectedrecovery period individuals as controls; We selected three tagSNPs (rs10415576,rs2304204and rs2304206)from HapMap Project in Chinese; Three SNPs in the IRF-3gene were genotyped by Multiplex SNaPshot technique. The genotype/allele frequenciesand Haplotype frequencies were calculated and analyzed.Results①The genotype frequencies for AA, GG and AG of rs10415576were35.23%,13.71%,51.07%in case samples, and40.48%,11.22%,48.30%in control samples. Thegenotype frequencies for AA, GG and AG of rs2304204were55.03%,5.48%,39.49%in case samples, and56.46%,4.76%,38.78%in control samples. The genotypefrequencies for CC, TT and CT of rs10415576were64.87%,3.05%,32.08%in casesamples and64.97%,2.38%,32.65%in control samples. No statistically significant difference between cases and controls (rs10415576, P=0.2105; rs2304204, P=0.8372;rs2304206, P=0.8315).②The allele frequencies for A, G of rs10415576were60.76%,39.24%in case samples,64.63%,35.37%in control samples. The allele frequencies for A, G of rs2304204were74.77%,25.23%in case samples,75.85%,24.15%in control samples. The allelefrequencies for C, T of rs230420were80.91%,19.09%in case samples,81.29%,18.71%in control samples. No statistically significant difference between cases and controls.rs10415576, P=0.0908, OR(95%CI)=1.1798(0.9740-1.4291); rs2304204, P=0.5959,OR(95%CI)=1.0579(0.8552-1.3133); rs2304206, P=0.8372,OR(95%CI)=1.0250(0.8097-1.2976).③The Haplotype frequencies for AAC, GAC, GGC and GGT of IRF-3were0.66%,14.11%,6.09%and19.04%in case samples;64.46%,11.39%,5.27%and18.71%incontrol samples. There were no statistically significant differences (AAC, P=0.0929;GAC, P=0.0916; GGC, P=0.4616; GGT, P=0.8642).Conclusion Our results suggest that the three SNPs sites in IRF-3were in strongLinkage Disequilibrium (LD), but no associated with HBV infection in Chinese Hanpopulation.
