Intervention Study On Acupuncture Combined With Buyang Huanwu Decoction On Cerebral Energy Metabolism Of Rats With Global Cerebral Ischemia

Objective:By detecting the acupuncture combined with Decoction on the indexes related to cerebral ischemia model of rat brain energy metabolism (ATP, ADP, AMP, TAN, EC) effects and pathological morphological structure, preliminary study on the effect on global cerebral ischemia rat brain energy metabolism mechanism of acupuncture combined with Decoction intervention.Methods:80 healthy SD rats were fed for 1 weeks, after 10 randomly selected as the blank group, the other rats were used to build model by four vascular occlusion method. Successful model of the rats were randomly selected 40 rabbits to divide into model group, decoction group, acupuncture and decoction group, positive drug group,10 rats in each group. After the success of modeling model group without intervention, the other three groups were subjected to the appropriate intervention, a total of 21 days of treatment. After the experiment, all of rats’brain tissue in each group were taken, and use to detecting the relevant indicators of brain energy metabolism (ATP, ADP, AMP, TAN, EC) and to observe the pathological morphological changes. The experimental results by using SPSS 18 software for statistical analysis.Results:The successful model of the rats in the intervention after treatment compared with the model group, acupuncture and decoction group, decoction group, positive medicine group could significantly increase the ATP, ADP content in brain tissue (P<0.01, P<0.05), TAN and EC value increased (P<0.01, P<0.05), and can significantly reduce the content of AMP in brain tissue (P <0.01, P<0.05), are obviously different; While the decoction group, acupuncture and decoction group, positive medicine comparison between the two two groups was no significant difference (P>0.05).2. The normal group rat hippocampus morphology of normal, arranged in neat rows, cells were spherical, clear edge, no edema, degeneration; model group rats hippocampal neuronal degeneration and necrosis, light microscope showed that the neurons in the nucleus pyknosis, smaller, triangular or conical, dyeing and deep, and arranged in disorder; and acupuncture and decoction group, decoction group, positive medicine group rat hippocampal neurons were improved in different level, and the acupuncture and decoction group improved most significantly.Conclusions:1. Ischemia rats after the obstacle is apparent in the energy metabolism of brain tissue, concrete embodiment in the production and utilization of ATP is greatly reduced, TAN and EC values are reduced. And this study shows that acupuncture combined with Buyang Huanwu Decoction can be made to improve the whole cerebral ischemia rats had energy metabolism, play the role of resistance to damage caused by cerebral ischemia.2. After intervention treatment observation of the rat hippocampus brain structure, the degree of clarity compared with model group, found that the treatment group of rat hippocampal neurons were different degrees of improvement, and acupuncture and decoction group works best.
