To detect the effects of new Alendronate-hydroxyapatite-chitosan (ALN/HA/CS) osteochondral composite scaffold materials on chondrogenesis differentiation, rabbit bone marrow-derived mesenchymal stem cells (BMSCs) were co-culture with the scaffold materials and the chondrogenic differentiation gene expression were researched in this experiment. Rabbit BMSCs were obtained by primary culture and subculture and co-cultured with ALN/HA/CS double-layer composite scaffold. The control group was double-layer composite material (HA/CS). In different stage, the cells were collected for real time quantitative PCR detection of ALP, collagen type Ⅱ and type I and glycosaminoglycans cartilage formation related gene expression and sugar production.Results:1 At 3,5,7 days, the ALP gene expression in the experimental group were significantly higher than the control group under osteogenic differentiation, the difference was statistically significant (P<0.05). At 7、14,21 day, the BGP gene expression in the experimental group were significantly higher than in the control group, the difference was statistically significant (P<0.05).2, At the 3,6,12 day, type II collagen gene expression and glycosaminoglycan (GAG) secretion volume was no significant difference between the experimental group and the control group under chondrogenesis culture. At the third day, type Ⅰ collagen gene expression was no significant difference between the experimental group and control group.At the 6、 12 day, type Ⅰ collagen gene expression levels in the experimental group were significantly lower than the control group, the difference was statistically significant (P<0.05).3, At the 3,6,12 day, type Ⅰ collagen gene expression levels in experimental group was not significantly different. In the control group, collagen-Ⅰ gene expression levels was increased at the 6 day than the 3 day, and increased at the 12 day than the 6 day.The difference was statistically significant (P <0.05).Conclusion:ALN/HA/CS cartilage composite material can effectively promote BMSCs into osteoblasts and inhibit cartilage phenotype degeneration of BMSCs into chondrocytes differentiation. ALN/HA/CS composite cartilage double-layer scaffold may be an ideal material for osteochondral repair.
