| ObjectiveThrough the observation of the effect of Tongdu Tiaoshen acupuncture group、routine acupuncture group 、 Tongxinluo drug group 、 model group on cerebral ischemia reperfusion model of neuronal apoptosis related proteins in rats caspase-9、caspase-3 expression level and the number of apoptosis cells. Comparison of three kinds of treatment methods of expression in model rat of caspase-9、caspase-3 on cerebral ischemia reperfusion and apoptosis cell count differences. And to investigate the mechanism and treatment advantage through Tong Du Tiao Shen acupuncture on cerebrovascular ischemic diseases affecting cell apoptosis.MethodChoose healthy adult male SD rats of clean grade, weighting for the 275±25g, through one week adaptive breeding, computer block randomly method to produce 24 rats served as sham operation group, fabricate model used Zea-Longe suture method, after duplication Zea-Longe neurobehavioral evaluation model is qualified or not, screening and retain qualified rats. Computer random number me divided into four groups, the model group, Tongxinluo drup group, routine acupuncture group and Tongdu Tiaoshen acupuncture group, 24 rats in each group, each group have different therapies. Strictly abide by the "3R" principle in the process of experiment.Routine acupuncture group selected Quchi, Hegu, Zusanli, Sanyinjiao acupoint; Tong Du Tiao Shen acupuncture group moxibustion Baihui 30 min every time, Dazhui prick bloodletting puncture every other day, 1-3 drops each time; Tongxinluo capsule for Tongxinluo drug group every time 1.0g/kg.d; model group rats after modeling successfully without any treatment; not inserted fishing line into the internal carotid artery for the sham operation group, the remaining processing congruence with model group. Each group of one times daily treatment, 7 days for one course of treatment, the processing is four courses. Beforetreatment and after completing treatment with Bederson method improved given neurobehavioral score. After final treatment,removed rats’ brains for specimen collection and detection for 24 h and 72 h.the results of comparative analysis used SPSS17.0 statistical analysis software for detection.Results(1) The nerve behavior score were significantly improved after successful replicated model for model group, Tongxinluo drug group, routine acupuncture group, Tong Du Tiao Shen acupuncture group.compared with sham operation group, neurological behaviour score of the four groups were very significant statistical significance(P<0.01),which showed that the MCAO/R model was successfully fabricated. And the model group, Tongxinluo drug group, routine acupuncture group, the Tong Du Tiaoshen acupuncture group mice through variance analysis neural behavior scores before treatment had no statistical significance(P>0.05). The results showed that the neural behavior scores of before and after treatment for model group、Tongxinluo drug group、routine acupuncture group、Tong Du Tiaoshen acupuncture group have comparability.(2) After the final treatment,Tongxinluo drug group, routine acupuncture group, Tong Du Tiaoshen acupuncture group give neurobehavioral score after treatment were obviously finished. Compared with the model group, the treatment groups differences in scores are very significant statistical significance(P<0.01); compared with the Tongxinluo drug group, routine acupuncture group and Tong Du Tiaoshen acupuncture group have very significant statistical significance in neurobehavioral score(P<0.01); compared with the routine acupuncture group, there is statistical significance through Tong Du Tiao Shen acupuncture group have significant statistical significance in neurobehavioral score(P<0.05),which indicate that Tong Du Tiao Shen acupuncture method better than Tongxinluo drug group and routine acupuncture group in reducing ischemia neurobehavioral score.(3) After the final treatment, model group, Tongxinluo drug group, routine acupuncture group and Tong Du Tiaoshen acupuncture group have different expression aboutcaspase-9 and caspase-3. Compared with the sham operation group, four groups’ expression of caspase-9 and caspase-3 were very significant statistical significance(P<0.01); compared with the model group, the relative expression was significant differences with three treatment groups(P<0.01, P<0.05); compared with the Tongxinluo drug group, the expression of routine acupuncture group and Tongdu Tiaoshen acupuncture group had statistically significant difference statistically significant(P<0.01, P<0.05); compared with the routine acupuncture group, the expression of Tongdu Tiaoshen acupuncture group has significant difference statistically significant(P<0.01, P<0.05).Within group comparisons, Tongdu Tiaoshen acupuncture group was statistically significant differences(P<0.05).The results showed that Tongxinluo drug, routine acupuncture,Tong Du Tiao Shen acupuncture method can reduce the expression of caspase-9、caspase-3 for ischemia reperfusion rats,Tong Du Tiao Shen acupuncture method effect is more significant compare with Tongxinluo drug group and routine acupuncture group.(4) After final treatment,the apoptosis cell number of model group, Tongxinluo d rug group, routine acupuncture group and Tong Du Tiao Shen acupuncture group w ere statistically significant difference. Compared with the sham operation group, t he apoptosis cell counts of four groups were very significant statistical significan ce(P<0.01); compared with the model group, the apoptosis cell counts of treatm ent groups were very significant statistical significance(P<0.01); compared with th e Tongxinluo drug group, apoptosis cell counts of routine acupuncture group and Tong Du Tiao Shen acupuncture group was significant differences(P<0.01, P<0.05); compared with the routine acupuncture group, the apoptosis cell counts of Tong Du Tiao Shen acupuncture group was statistically significant differences(P<0.05). Within group comparisons, Tongdu Tiaoshen acupuncture group was statistically s ignificant differences(P<0.01).The results showed that Tongxinluo drug, routine ac upuncture,and the Tong Du Tiaoshen acupuncture can reduce the number of apopt osis cell for ischemia reperfusion rats, Tong Du Tiao Shen acupuncture method effect is more significant compare with Tongxinluo drug group and routine acupunc ture group.Conclusion(1)Tongxinluo drug, routine acupuncture and Tong Du Tiao Shen acupuncture can adjust the score of on cerebral ischemia reperfusion neurobehavioral model rats obviously.Routine acupuncture is superior to Tongxinluo drug.Tong Du Tiao Shen acupuncture is better than that of routine acupuncture,so the effect of Tongdu Tiaoshen acupuncture comparing with Tongxinluo drug and routine acupuncture on cerebral ischemia reperfusion neurobehavioral score were significant for the ischemia reperfusion model rats.(2)Tongxinluo drug,routine acupuncture method and Tongdu Tiaoshen acupuncture can effectively reduce expression and the number of apoptotic cell of the caspase-3, caspase-9 on ischemia reperfusion rat.Routine acupuncture is better than Tongxinluo drug, and Tongdu Tiaoshen acupuncture is better than the routine acupuncture,which show that Tongdu Tiaoshen acupuncture is better than that of intervention of Tongxinluo drug and routine acupuncture.(3)Through the Tongdu Tiaoshen acupuncture effectively mend cerebral ischemia reperfusion harm for the ischemia reperfusion model rats.It may be associated with decreasing the expression of caspase-9 and caspase-3, restraining apoptotic cell, thus forming cerebral protection. |
