Objective: By observing the expressions of Endostatin(ES)phosphorylation and caspase-9 in the hippocampus of rats after cerebral ischemia reperfusion injury induced by acupuncture at Dazhui,Baihui and Renzhong,the brain protective effect of acupuncture on cerebral ischemia reperfusion injury and its possible mechanism were discussed.Methods: Middle cerebral artery ischemia(MCAO)model was prepared by using Zea Longa occlusion method.120 male SD rats were randomly divided into the sham operation group,the model group,the drug group,and the acupuncture point group,with 30 rats/group.After the success of the building,the control group,model group,only drug group in 3 mg/Kg(diluted to 1 ml)intraperitoneal injection,acupuncture and group does great job,the people,the best will point acupuncture,12 h/times,for the first time before and 6 times to complete treatment after nerve function defect grade,TTC staining to observe rat ischemic cerebral infarction area,Western Blot method to detect ES phosphorylation expression;The expression of caspase-9 was detected by immunohistochemistry.Results: 1 Comparison of neurological deflicit scores: comparison between groups,before treatment,there were statistically significant differences between the model group,the drug group and the acupuncture point group(P < 0.01);There was no statistically significant difference between the 3 model groups(P>0.05).After treatment,there were statistically significant differences among the model group,drug group and acupuncture point group(P < 0.01).2 Comparison of the ratio of cerebral infarction area: after treatment,compared with the sham operation group,the ratio of cerebral infarction area in the model group,acupuncture point group and drug group increased,and the difference was statistically significant(P<0.01).Compared with the model group,the acupuncture point group showed statistically significant differences(P < 0.01).3 Comparison of ES: After treatment,there were statistically significant differences between the model group,drug group and acupuncture point group(P < 0.01),compared with the model group,there were statistically significant differences between the drug group and the acupuncture point group(P < 0.05).4 Comparison of Caspase-9: After treatment,there were statistically significant differences between the model group,drug group and acupuncture point group(P<0.01).Compared with the model group,there were statistically significant differences between the drug group and the acupuncture point group(P<0.01).Compared with the drug group,the acupuncture point group showed statistically significant differences(P<0.01).Conclusion : 1.Acupuncture can reduce the score of nerve function defect and the ratio of cerebral infarction area in rats with cerebral ischemia reperfusion injury,and realize the brain protection effect.2.Acupuncture can reduce the risk of cerebral ischemia reperfusion injury,and its effect mechanism may be related to the down-regulation of ES expression and the promotion of angiogenesis.3.Acupuncture can down-regulate the expression of caspase-9 in the side of cerebral ischemia and reduce cerebral ischemia reperfusion injury,which is related to the fact that acupuncture can reduce the production of apoptosis factors.4.The expression of ES was positively correlated with the expression of caspase-9,and the anti-ischemia reperfusion injury mechanism of acupuncture may be related to the regulation of caspase-9 expression by ES,thereby inhibiting apoptosis.
