The Influence Of The Electro Acupuncture On Du Meridian Acupoints To The Expression Of The Ability Of Learning And Memory Of Rats With Vascular Dementia,Which Also Have An Effect On Hippocampus’s VEGF And FLT-1,FLK-1MRNA Receptor Expression

Objective:The influence of the acusector on Du meridian acupoints to the expression of the ability of learning and memory of rats with vascular dementia,the key factor of the hippocampus promoting angiogenesis VEGF and its receptor fms-like Tyrosine kinase-1,Vascular endothelial growth factor receptor 2 was observed.Further discussion about the mechanism of acusector treating vascular dementia from the aspect of angiogenesis and theoretical and experimental evidence basis for treating vascular dementia on Du Meridian acupoints will be provided.Methods:(1)110 clean adult male Wistar rats were elected.After adaptive feeding,14 rats were produced as sham operation group which was adopted an random number table method while the rest ones were adopted Pulsinelli’s vascular dementia 4-vessle occlusion model that has been modified.On the seventh day after modeling,and then Morris water maze test was done on survival rats for one day.The reference value was set as the mean of the average escape latency period of rats in sham operation group.The proportion of every rat’s difference between average escape latency period and reference value account for the time of average escape latency period in operation group were measured. The value more than 20% is defined as dementia rats.(2)The successful model rats were randomly divided into acusector group,model control group and Western medicine group.On the eighth day after surgery,the acusector group and the Western medicine group were given acupuncture and Western medicine treatment,respectively.The treatment consisted of two courses in total while each course of treatment consisted of ten times and one time per day.Acusector treatment group were paralleled with sham operation group and model controlled group which fixed rats on the rat plates only and the courses of treatment were the same as the acusector group and the Western medicine group.After two courses of treatment,Morris water maze test should been done for five days.Place navigation test was done in the former four days which was designed to record the average escape latency period.The spatial search testwas done on the fifth day which was designed to record the numbers of rats across the original platform and proportion of swimming time and distance in the original platform quadrant account for entire swimming time and distance.The materials were taken after Morris detection of water maze accomplishing.Verify that the success of rats after modeling and the changes of learning and memory abilities of rats in each group were tested.The treatment effect of acusector on Du meridian points on vascular dementia was evaluated.(3)VEGF in the hippocampus and the expression of its receptor Flt-1、Flk-1 m RNA of rats in each group were tested by using RT-PCR.Whether mechanism of acusector on Du meridian points to VD rats is related to angiogenesis was further judged.Results:(1)results of place navigation test:The difference about the escape latency time between four groups had statistically significance(P<0.01).Comparing with the sham operation group,the learning ability of rats in the model control group was decreased obviously(P<0.01).Comparing with the model control group,the latency period of rats in acusector group and Western medicine group was improved obviously(P<0.01).The difference of academic record in rats between acusector group and Western medicine group had statistically significance(P<0.01).(2)results of spatial search test:The time of crossing the platform of rats in model control group were less than the rats in sham operation group,acusector group and Western medicine group(P<0.01 or P<0.05).The acusector group had not reached the level basically with sham operation group(P<0.01) which difference had statistically significance between the two groups(P<0.01).In aspect of time of spatial search,the rats in model control group were worse than the ones in sham operation group,acusector group and Western medicine group(P<0.01 or P<0.05).The acusector group had no statistically significance with sham operation group and Western medicine group(P>0.05) while it had statistically significance between Western medicine group and sham operation group(P>0.05).In aspect of distance of spatial search,the rats in sham operationgroup,acusector group and Western medicine group were better obviously than the ones in model control group(P<0.01 or P<0.05).But acusector group and Western medicine group were still worse than sham operation group(P<0.01 or P<0.05) while it had no statistically significance between the former two groups(P>0.05).(3)Zea Longa score of neurological behavior of rats in each group:Zea Longa score of neurological behavior in electric acupuncture group, model control group and Western medicine group were still higher than that in sham operation group after treatment(P<0.01)while Zea Longa score of neurological behavior in electric acupuncture group and Western medicine group were lower than that in model control group obviously(P<0.01).The comparison between electric acupuncture group and Western medicine group had statistically significance(P<0.01).The behavioral score indicate that electric acupuncture on Du meridian acupoints can decrease the score of neurological behavior in VD rats obviously.(4)RT-PCR testing result:Comparing with sham operation group, hippocam-pus VEGF m RNA、Flt-1m RNA、Flk-1m RNA expression increased obviously in model control group,acusector group and Western medicine group which all had statistically significance(P<0.01).After acusector on Du meridian points aniracetam,expression of hippocampus VEGF m RNA had increased obviously comparing with model control group(P<0.01).The difference had no statistically significance between model control group and Western medicine group(P>0.05).
