Objective Periodontitis, trauma, cancer and congenital malformations can cause varying degrees of defect, has been a difficult clinical treatment, Farly autologous bone graft has been regarded as the gold standard, but in order to reduce the secondary trauma, gradually switch to allogeneic bone graft material, it is expensive, and the presence of infection and antigenicity and other issues. So in recent years for bone tissue engineering is gradually attention. Scaffolds as seed cells and growth factors carrier, plays a vital role. Not only imitate the simple pursuit of the structure and composition of bone, which requires scaffolds have good organizational compatibility, biodegradability, osteoinductive and osteoconductive properties. By appropriate choice of scaffold suitable manufacturing process to produce a composite scaffolds meet the above characteristics, is become a hotspot to the bone tissue engineering research.Bletilla striata main chemical components is Bletilla striata glucomannan, BSG. Has been used in traditional Chinese medicine for many years. Because Bletilla striata has anti-inflammatory and anti-tumor properties, have a good biocompatibility and biodegradability, can be used in various forms of drug delivery and bioremediation materials. Chitosan (Chitosan, CS) is the only natural polysaccharide basic polysaccharide. CS can be decomposed and absorbed by the body through the body lysozyme. Because of its rich source, non-toxic, good biocompatibility and prevent drug diffusion and dissolution and other characteristics, can be well used in drug carrier. But CS-soluble poor, only soluble in acidic solution, insoluble in water and many organic solvents, largely limits its application. The CS hydroxypropylation prepared water-soluble hydroxypropyl chitosan (hydroxypropyl chitosan, HPCS), not only retains the CS basicity and positive electricity, while also improving the performance of CS can be directly dissolved in water, no need to dissolve in acidic solvent, nacre is the innermost part of the shell, surrounded by organic matter constitutes bracket aragonite calcium carbonate. Many studies have confirmed its structure and mechanical strength similar to cortical bone, nacre powder has good biocompatibility, bioactivity and biodegradability, and a bone conduction and bone induction effect. As a new alternative biomaterials for bone repair, extensive research by scholars.Our previous research has its own three-dimensional composite of nano-nacre powder, NNP, hydroxypropyl chitosan, HPCS and dialdehyde Bletilla striata glucomannan, DBsGM consisting of scaffold, and the experiment proved its good physical properties, biocompatibility and biodegradability. This experiment will applied NNP/HPCS/DBsGM porous scaffolds in rat mandibular defect, provide experimental evidence for the clinical application of the composite scaffold.Methods 32 male Wistar rats, each animal with bilateral mandibular molar area box defects, according to the disposal of time (2,4,6,8 weeks) were randomly divided into 4 groups:each group of eight, four groups of the experimental side were implanted with letilla striata glucomanna/hydroxypropyl chitosan/nano-nacre powder composited scaffolds.
