Objective: Through observing the expression of tyrosine kinase receptor B(TrkB) and brain derived neurotrophic factor(BDNF) in papillary thyroid carcinoma(PTC), comparing the difference expression with clinical data, exploring the correlation each other and the relationship with occurrence and development of papillary thyroid carcinoma, thus providing a theoretical basis for clinical application.Methods: To gather thyroid tissue specimens with completed clinical data and pathologically confirmed from Hunan Provincial People’s Hospital January 2012 to January 2014, including 48 cases of papillary thyroid carcinoma, 46 cases of thyroid adenoma, 46 cases of normal thyroid tissue, 22 cases of nodular goiter. Immunohistochemical streptavidin was used to detect the expression of TrkB and BDNF.Results: 1.TrkB and BDNF protein expression in thyroid adenoma and papillary thyroid carcinoma,no expression in normal and thyroid nodular goiter; TrkB and BDNF protein expression was mainly localized in the cytoplasm and cell membrane of thyroid tissue.2.TrkB and BDNF protein expression in thyroid adenoma and papillary thyroid carcinoma was sign- ificantly higher than normal thyroid tissue(P<0.05); Trk B and BDNF protein expression was no significant difference in the nodular goiter and normal thyroid gland(P>0.05);TrkB and BDNF protein expression in the thyroid adenoma and papillary thyroid carcinoma was significantly higher than nodular goiter(P<0.05); Trk B and BDNF protein expression was no significant difference in thyroid adenoma and papillary thyroid carcinoma(P>0.05).3.TrkB protein expression in papillary thyroid carcinoma was correlated with tumor size, lymph node metastasis, differentiation and clinical stage(P < 0.05), without the gender and age(P > 0.05).4.BDNF protein expression was correlated with tumor size, differentiation(P<0.05), without age, gender, lymph node metastasis and clinical stage(P>0.05).5.TrkB and BDNF protein expression in papillary thyroid carcinoma was positive correlated.Conclusion: 1)TrkB protein expression in the papillary thyroid carcinoma was significantly higher than normal thyroid tissue and thyroid nodular goiter, suggesting TrkB may play an important role in occurrence of papillary thyroid carcinoma.2)BDNF protein expression in the papillary thyroid carcinoma was significantly higher than normal thyroid tissue and thyroid nodular goiter, suggesting BDNF may play an important role in occurrence of papillary thyroid carcinoma.3)TrkB protein expression in papillary thyroid carcinoma was correlated with tumor size, lymph node metastasis, differentiation and clinical stage, suggesting TrkB may affected the development of papillary thyroid carcinoma. 4)BDNF protein expression was correlated with tumor size, differentiation and clinical stage, suggesting BDNF may affected the development of papillary thyroid carcinoma. 5) TrkB and BDNF protein expression in papillary thyroid carcinoma was positive correlated, suggesting that they may synergies and through a common signaling pathway to mediate the occurrence and development of papillary thyroid carcinoma.
