Objective:Detection of panax notoginseng on lower limb ischemia rat model of vascular endothelial growth factor(VEGF), the capillary density(MVD) and CD34+, FLK-1+ and blood enzyme and the effect of notoginseng on the angiogenesis of lower limbs ischemia in rats, looking for effective method for clinical treatment of lower limb ischemic diseases. Methods:180 SPF SD rats randomly grouped into: normal group, model group, notoginseng, high dose group, the three dose groups, notoginseng in low dose group, the effect; Model group, notoginseng and the effect of group line on the left side of the lower limb artery ligation in rats and branch out, result in lower limb vascular ischemia model. Notoginseng including dose groups, respectively, in accordance with the 0.572 g/kg, 0.286 g/kg, 0.143 g/kg for postoperative lavage; Effect was set according to 0.01 g/kg irrigation notoginseng; 1 times a day, for 14 consecutive days. Normal group and model group to fill the distilled water.Respectively in 3 days, 7 days, 14 days after random take each group 10 rats, peripheral blood creatine kinase(CK), lactate dehydrogenase(LDH), aspertate aminotransferase(AST), the determination of CD34+, FLK-1+; Take lateral gastrocnemius, observe the capillary structure of lower limbs ischemia recovery, calculating capillary density(unit vision aizen area/unit field area); The left leg hematoxylin- eosin(HE) pathological dyeing, observation number between skeletal muscle cells in the blood vessels; Determination of VEGF protein expression in muscle tissue. Postoperative day 14 groups of left lower limb necrosis of cases. Results:1.Three days postoperatively, ischemia rats hind limbs temperature is reduced, the color white, not seen gangrene, wound scab over, did not see obvious hemorrhage, 7 days after wound healing, 14 days after the scabs fall off, see model group 1 case of ischemic hind legs black foot pad, 1 nail to fall off.2.Blood enzyme determination: normal group blood enzyme value every time there was no significant difference; More than 3 days postoperatively, set each AST, CK, LDH values are higher than blank group, model group, notoginseng, low dose group of AST, CK value notoginseng high, middle dose group and the effect was significantly higher(P < 0.01); After 7 days, each AST, CK, LDH values drop, notoginseng, high, medium and low dose group and the effect of group compared with model group decreased significantly(P < 0.05), notoginseng, high dose group of the most significant(P < 0.01); 14 days after surgery, the AST, CK, LDH values from the previous trends to slow down, and no obvious difference with 7 days postoperatively, and 3 days after significant difference(P < 0.05).3. CD34+ and FLK-1+ positive cells: 3 days postoperatively, notoginseng, high dose, express to rise the effect compared with model group, significant difference(P < 0.01), notoginseng high-dose group increased most significantly; 7 days postoperatively, notoginseng high dose a significant rise in proportion than other groups(P < 0.05); After 14 days, the expression is reduced.4. The determination of VEGF expression: 3 days postoperatively, notoginseng, high, medium dose, express to rise the effect compared with model group, significant difference(P < 0.05), notoginseng, high dose group of VEGF protein expression values the highest; 7 days after each VEGF expression began to decline, notoginseng, high, middle dose and effect group decreased significantly(P < 0.01); 14 days postoperatively, notoginseng, high dose group of VEGF expression increased significantly in the other group(P < 0.05), notoginseng, high dose group, the three dose groups, the effect in group a expression to reduce postoperative 3 days significant difference(P < 0.05).5. Determination of capillary density(MVD) : 7 days postoperatively, notoginseng, high dose group had a significantly higher MVD value model group(P < 0.05); 14 days postoperatively, notoginseng, high dose group had a significantly higher MVD value model group and low dose group(P < 0.05).6. The left leg hematoxylin- eosin(HE) pathological dyeing: neat rows blank group rats muscle cells, muscle fibers have a certain amount between vascular distribution; Model group rats with muscle atrophy and interstitial hyperplasia, vascular number increased in the blank group; Notoginseng high dose group of muscle atrophy in rats is lighter, vascular distribution between muscle fiber number more than model group; Three, low dose group of muscle fibers in blood vessels between populations, but not as good as high dose group; Group effect was slightly increased vascular distribution. Conclusion:Through determination of each index of rat, a preliminary conclusion notoginseng in lower limb ischemia rat model can promote VEGF expression in ischemia group, make the CD34+, FLK-1+ positive cells increased, thus promoting angiogenesis, reduced state of limb ischemia. The effect of high dose group is better than the low dose group, it may be able to mobilize endothelial progenitor cells(epcs) and panax notoginseng and make it to the relevant migration and promote its proliferation differentiation of ischemic tissues.
