In this thesis work,the cell model of inflammatory human endometrium was built up,and the most proper Concentration of effective components of panax notoginseng and gongxiulin (panax notoginseng saponines 12.5mg/ml and dencichen 3.13mg/ml; gongxiulin 5mg/ml) was determined by the MTT assay of effective Components of panax notoginseng and gongxiulin on the cells of endometrium; and stopping bleeding and restoring of infection-induced human endometrium was determined by the NF-KB,TNF-α,IL-1.e results show:1,Establishment of Cell Model of Inflammatory Human EndometriumObjective:To establish the cell model of inflammatory human endometriun by the lipopolysacharide. Methods:The effect of five concentration grades of lipopolysaccharide on endometrium cells was investigated by the MTT assy. The optimized concentration, whose inhibit rate(IR)to the growth of endometrium cells was less than or epual to 10 percent, was selected to setup cell models. Then concentration variation of interleukin 1β(IL-1β) and tumor necrosis factor-α(TNF-α) of endometrium cells in the control group, the 48h model group and the 72h model group were recorded. Result:100μg/ml is the optimized concentration of lipopolysaccharide by the standard that IR is less than or epual to 10 percent. The concentrations of IL-1βand TNF-αof the 48h model group and the 72h model group are much higher than the control group's, which proves the successful establishment of the cell model of inflamma tory human endometriun by the 100μg/ml lipopolysaccharide. Conclusion:100μg/ml is the optimized concentration of lipopolysaccharide to setup the cell model of inflammatory human endometriun.2,The selection of the optimized concentration of the effective components of panax notoginseng and gongxiulinObjective:To select the optimized concentration of the effective components of panax notoginseng and gongxiuin to endometrium cells. Conclusion:The concentration of the panax notoginseng saponines 12.5mg/ml and the dencichen 3.13mg/m; 1 gongxiulin 5mg/ml is the optimized concentration of the effective components of panax notoginseng to be applied in the subsepuent research.3,Influence of two kinds of active components from Panax notoginseng on NF-KB activity and TNF-αas well as IL-1 mRNA expression in inflammatory metrorrhagia model of Macaca mulatta.Objective:To explore the changes of NF-KB activity and TNF-αas well as IL-1 expression in the infection-induced metrorrhagia model of macapue, influence of two kinds of active components from Panax notoginseng on NF-KB activity and TNF-αas well as IL-1 Mrna expression.Methods:Electrophoretic mobility shift assay (EMSA)was performed on the nuclear extracts to detect the activity of NF-KB and RT-PCR was performed to detect the express of TNF-α,IL-1mRNA in three groups. Result:compared to model group,the NF-KB activity and TNF-α, IL-1mRNA expression of TCM group was weaker. The NF-KB activity TNF-α, IL-1mRNA expression of TCM group is similar to that of normal group. Conculusion:The activation of NF-KB and the over-expression of TNF-αIL-1mRNA Participate in the pathognesis of infection-induced metrorrhagia in human endometrium.The reason two Kinds of active components from panax notginseng decrease metrorrhagia is perhaps that it can inhibit the activation of NF-KB and expression of TNF-α,IL-1mRNA.
