Background and Objectives The point Danzhong is specific for respiratory system, cardiac system and so on. The anatomical location of the point Danzhong and the part of chest compression of standard cardio-pulmonary resuscitation (CPR) are coincided. In this study, we used electro-acupuncture (EA) to stimulate the point Danzhong, and combined with standard CPR. Then we rescued the rats of cardiac arrest. The rates of recovery of automatic circulation and successful resuscitation were observed. And the blood pressure and left ventricular function were also observed. Then we analyzed the function of EA stimulation with the point Danzhong for CPR.Methods The study was divided into two parts. The first part was to find what different influence on blood pressure and left ventricular function with different frequencies of EA to stimulate the point Danzhong in rats.20 SD rats were randomly divided into four groups with 5 rats in each group, namely the control group,20Hz EA group,50Hz EA group and 100Hz EA group. The control group was without EA stimulation, and was only given surgical procedures (including electrocardiograph monitoring, arterial and venous catheters). The rest of the rats after surgical operations, were given the EA stimulation of different frequencies, and continued 5min. The beginning of the EA stimulation was recorded as Omin. We recorded the blood pressure of each group before the EA stimulation, and at Omin,5min, 10min,30min,60min, as well as the echocardiographic images in 1h.The second part of the study was to explore the effectiveness of CPR by stimulating the point Danzhong with EA in rats with cardiac arrest.100 SD rats were randomly divided into four groups, namely control group, standard CPR group, EA with CPR group and EA before CPR group. Each of group were 25 rats. The control group did not induce cardiac arrest and resuscitation, just given the surgical procedures (including electrocardiograph monitoring, endotracheal intubation, arterial and venous catheters). Other groups were induced cardiac arrest. After cardiac arrest, the standard CPR group observed 5min (intermission), then followed by standard CPR, including chest compression, mechanical ventilation and medications of rescue. EA with CPR group were observed 5min after cardiac arrest, then given EA stimulation with the point Danzhong while given standard CPR. The time of EA stimulation was 5min. EA before CPR group stimulated the point Danzhong with EA immediately after arrest. The time of stimulation was also 5min. If the rats were not restored spontaneous circulation after 5min, we would give the standard CPR immediately. We recorded the rates of ROSC and successful recovery. And the time of ROSC was regarded as Omin. We recorded the blood pressure before the EA stimulation and the pressure at Omin,5min, 10min,30min, and the echocardiographic images at 1h.Results The first part was the different effects on blood pressure and left ventricular function with different frequencies EA stimulation with the point Danzhong. The influence of the frequency with 20Hz was not obvious. The frequency with 50Hz can significantly improve the blood pressure at 5min and 10min. And the difference with control group was statistically significant (117.0±12.6vs.86.2± 12.2,P=0.007;116.0±8.0vs.85.4±12.5,P=0.007). Meanwhile 50Hz can also improve left ventricular ejection fraction (LVEF) and left ventricular fractional shortening (FS). And there were significant differences between the control group (91.0±1.46vs.79.9± 3.4,P=0.000;56.5±2.4vs.42.7±3.1,P=0.000).100Hz has little effect on blood pressure. But the LVEF and FS were significantly lower than control group (3.7±2.9vs.79.9± 3.4,P=0.005;37.5±2.7vs.42.7±3.1, P=0.017).The second part of study was to find the function of the EA with the point the point Danzhong for CPR. The rate of ROSC of EA with CPR group (72%) was higher than standard CPR group (40%). The difference was statistically significant (x 2=5.195,P=0.023). The rate of successful recovery (40%) compared with standard CPR group (28%) showed no difference (x 2=0.802,P=0.370). The blood pressure of EA with CPR group was lower than standard CPR group at Omin (139.9±3.9vs.152.0± 7.5,P=0.027), while it was higher at 30min (85.1±4.3vs.70.4±6.4,P=0.001). And the LVEF and FS values were higher than standard CPR group. The difference was statistically significant (82.3±3.7vs.68.5±4.8,P=0.000;45.7±3.5vs.33.0± 3.3,P=0.000). Compared with standard CPR, the rate of ROSC and successful recovery of the EA before CPR group was increased significantly (80%vs.40%, x2=8.333, P=0.004;60%vs.28%, x 2=5.195, P=0.023). The blood pressure of the EA before CPR group at Omin was lower than standard CPR. The difference was significant (140.6± 12.8vs.152.0±7.5,P=0.040). But at 30min, the blood pressure was higher than standard CPR group with statistical significance (85.9±8.9vs.70.4±6.4,P=0.01). In EA before CPR group, the rats of successful resuscitation without chest compression was 8 rats, and was higher than standard CPR group which was 2 rats. The difference was significant (x 2=4.500,P=0.034). And the time of compression was shorter than standard CPR group (2.1±0.6vs.3.8± 1.3,P=0.015). The LVEF and FS were also higher than standard CPR group. There were significant differences (76.9±2.9vs.68.5 ±4.9,P=0.001;39.9±2.7vs.33.0±3.3,P=0.002).Conclusions 1.EA stimulation with the point Danzhong at different frequencies are different on blood pressure and left ventricular function in rats.50Hz can improve blood pressure and enhance left ventricular function. But 100Hz reduces cardiac function.2. Stimulation with EA on the point Danzhong can increase the effective role of CPR. CPR with EA stimulation, even before or during, can improve the rates of ROSC and success resuscitation of cardiac arrest of rats. The influence of EA before CPR is obvious. And EA stimulation with the point Danzhong can improve the blood pressure at 30min after ROSC, and can increase the left ventricular function. And the effect of EA stimulation with CPR is more advanced on left ventricular function.
