Objective: Research on CD4+CD25+ regulatory T cells(CD4+CD25+ Treg), serum 25- hydroxyvitamin D(25-(OH)D3) and different expression of serum total Ig E, specific Ig E in 1-5 years old children with wheezing in peripheral blood, to explore CD4+CD25+ Treg, 25-(OH) D3 and serum total Ig E and specificity Ig E in clinical significance in children with asthmatic diseases, to provide new ideas for clinical treatment of children with asthmatic diseases.Methods: Choose the Third Affiliated Hospital of Soochow University Department of Pediatrics in June 2014- December 2014, those has not been used glucocorticoid treated in hospital with wheezing symptoms in children 74 cases.According to the presence or absence of atopic dermatitis was diagnosed in the past, allergic rhinitis or family history of asthma group, with one of the above were classified as special recruitment group(40 cases), do not have candidates who are classified as non-specific group(34 cases). And select the same period in surgical in-patient treatment of inguinal hernia, kidney stones and other non inflammatory diseases of children in 20 cases as normal control group. The two groups were on the day of admission from peripheral venous blood, using measured by flow cytometry in peripheral blood CD4+CD25+ Treg, CD4+T cells and CD8+T cells, NK cells and T cells and B cells, enzyme-linked immunosorbent assay(ELISA) in detection of serum vitamin D active metabolite 25-(OH) D3 level, Western blot method for determination of serum total Ig E and specific Ig E levels.Results:(1) Compared the normal control group, the peripheral blood CD4+CD25+ Treg(7.71±2.45) %, to the asthmatic group of peripheral blood CD4+CD25+ Treg(5.81±2.84) % significantly reduced(P<0.05). Asthmatic group(special recruitment group and non-specific group) within group comparison shows, special recruitment group of peripheral blood CD4+CD25+ Treg(5. 18±3.17) % lower than non-specific group(6.55+2.23) %, the difference was statistically significant(P< 0.05).(2) The normal control group of peripheral blood CD4+ T cells and CD8+ T cells, NK cells, T cells and B cells were(22.13±5.79) %,(34.53±5.42) %,(9.92±2.95) %,(65.84±4.98) %,(20.87±4.06) %, compared with the normal control group, asthmatic group of CD8+ T cells(29.51±8.43) %, T cells(60.76±9.48) % significantly decreased(P< 0.05), B cells(26.86±8.73) % was significantly increased(P< 0.05), while CD4+ T cells, NK cells there was no significant difference between the two groups(P> 0.05). Asthmatic group(special recruitment group and non-specific group) within group comparison shows, two groups of peripheral blood CD4+ T cells and CD8+ T cells, NK cells and T cells and B cells showed no significant difference(P> 0.05).(3) Compared with normal control group, asthmatic group, the serum 25(OH) D3 level is reduced, statistically significant difference(P < 0.05). While in the asthmatic group(special recruitment group and non-specific group) within group comparisons showed no significant difference(P > 0.05).(4) Asthmatic group(special recruitment group and non-specific group)within group comparisons, especially for the special recruitment group serum specific Ig E was significantly higher than that of non-specific group, the difference was statistically significant(P < 0.05); Serum total Ig E in special recruitment group was significantly higher than that in non-specific group(P < 0.001).Conclusion: 1. Peripheral blood CD4+ CD25+ Treg, serum 25(OH) D3 level decreased and the cellular and humoral immune regulating function disorder, may be associated with asthmatic disease are related to.2. The proportion of peripheral blood CD4+ CD25+ Treg and serum 25(OH) D3 level rise is a part of the body’s immune defense performance, in particular, apply to those who with the atopy physique of children with wheezing, peripheral blood CD4+ CD25+ Treg can be used as new targets of its breathing early treatment.
