Objective Parkinson disease is common in the elderly progressive disese of central nervous system, is pathologically characterized are the progressive loss of dopaminergic neurons in the substantia nigra pars compacta. So far, the causes and mechanisms of Parkinson’s disease is unclear. The present study suggest that the incidence of Parkinson’s disease and environmental factors, genetic factors, nervous system aging and a variety of factors related to the interaction. The disease by British physician James Parkinson(James Parkinson) was first reported in 1817.1. To observe number and apoptosis of dopaminergic neurons in mouse model of parkinsonism induced by little dose of(MPTP)injection.2. To observe expression of microglia and behavioral changes in chronic mouse model of parkinsonism induced by little dose of(MPTP)injection.3. To observe expression of inflammatory cytokines TNF-αand IL-1βin chronic mouse model of parkinsonism induced by little dose of(MPTP)injection.Methods This investigation adopted C57BL/6 model of chronic parkinsonism induced by little dose of MPTP injection, set test group and normal control group:16 month C57BL/6 male mice(n=40)were divided into the control group and MPTP model group, with model group was divided into1、3、5weeks, each group was received 15mg/kg MPTP intraperitoneal injection, 2 times a week, the behavioral was observed when the injection was completed. Each model group was unified drawn after 48 h. Immunohistochemistry analyses microgliaand TH in the substantia nigra striatum.Results The findings of this study were as follows:1.Behavior observation results: compared with the control group, the model group of mice coordination was decreased, and erected hair, front legs elevated, began to appear restless, running back and forth in the cage, slight tremors, bow curled back into the group,nodding disease, and then appeared briefly reduced activity, response to external stimuli.2.Morphologic examination results: HE staining found that normal neuron aggregation, deeply stained nuclei, neuronal size and shape differences, their cell bodies are round, spindle and cone-shaped.3.Microglia immunohistochemical results : Compared with the control group, a week model group, positive microglia has a growing trend, a slight enhancement of the immune response( P > 0.05).Three weeks, positive microglia increased significantly,but the majority of small cell body, protruding more slender; after five weeks, there are more positive microglia, cell body larger, protruding shorter, immune respone stronger, showed significant active state, compared with the control group, there were significant statistical significance(P<0.05).4.The expression of TNF-α和 IL-1βin substantia nigra in experimental group Showed a gradual increasing trend,and the 1week,there was no significant difference compared with the control group.Conclusions MPTP has a distinct impairment on motor function of C57BL/6 mice. MPTP induced C57BL/6 mouse to appear parkinsonism symptom and microglia was activated in striatum nigra system and secrete large amounts of inflammatory cytokines. Microglia participate in MPTP-induced chronic inflammation in the brain, and involved in the occurrence and development of the Parkinson disease.
