Objective: This work aimed to evaluate the expression of RYBP, ZEB1 and ZEB2 in hepatocellular carcinoma tissues and adjacent noncancerous tissues, to investigate their correlation with the clinicopathological features and clinical prognosis, and to preliminarily discuss their relevance in carcinogenesis and development of HCC. Methods : Immunohistochemical technique was used to detect the expression of RYBP, ZEB1 and ZEB2 on tissue microarray slice obtained from 216 cases of hepatocellular carcinoma tissues and 216 cases of adjacent noncancerous tissues, to investigate their correlation with the clinicopathological features and their relevance; Western blotting technique was used to detect the expression of RYBP from 30 cases of hepatocellular carcinoma tissues and 30 cases of adjacent noncancerous tissues; Western blotting technique was used to detect the expression of RYBP from HCC cell lines and normal liver cell lines,as well. Results:(1)The positive expression rate of RYBP in HCC tissues was significantly lower than that in adjacent noncancerous tissues; The expression of RYBP in HCC tissues was related to tumor diameter and metastasis or not;The negative expression of RYBP was significantly related to poor prognosis of HCC patients, and as RYBP expression level was increased gradually, the patients’ survival time was significantly prolonged.(2)RYBP protein expression level in HCC tissues was significantly lower than adjacent noncancerous tissues by detecting RYBP protein expression in 30 cases of hepatocellular carcinoma tissues and adjacent noncancerous tissues; RYBP protein expression level was down-regulated in Chang Liver,PLC and Huh7 cells,and was increased in L02, HepG2 and SMMC7721 by detecting RYBP protein expression in HCC cell lines and normal liver cell lines.(3) The positive expression rate of ZEB1 and ZEB2 in HCC tissues was significantly higher than that in adjacent noncancerous tissues and the expression of ZEB1 and ZEB2 in HCC tissues was related to metastases or not;The positive expression of ZEB1 and ZEB2 was significantly related to poor prognosis of HCC patients, and as ZEB1 and ZEB2 expression level was decreased gradually, the patients’ survival time was significantly prolonged.(4) The expression of PYBP has a negative correlation with the expression of ZEB1 and ZEB2. Conclusions:(1) The negative expression of RYBP promoted HCC invasion and metastasis, and played a role in carcinogenesis and development of HCC, and had certain clinical value to judge the prognosis of patients with HCC.(2) It futher validated that ZEB1 and ZEB2 participated in carcinogenesis and development of HCC involved EMT, and they had intimate connection with the invasion and metastasis of HCC, and they were important indicators to estimate the prognosis of HCC patients.(3)The expression of RYBP had a negative correlation with the expression of ZEB1 and ZEB2 in HCC, and it predicted that RYBP might participate in HCC EMT progress.
