PurposeThis experiment adopts the method of jiaji electroacupuncture combined with treadmill training to explore effects on bone strength and bone mineral density of tibia of rabbit with sciatic nerve injury. In older to discuss the effectiveness that jiaji electroacupuncture combined with treadmill training improve the bone metabolism after peripheral nerve injury.Methods24 New Zealand rabbits were randomly divided into four groups:model group, treadmill group, sham group, jiaji electroacupuncture combined with treadmill group, there were 6 New Zealand rabbits in each group。The sciatic nerve of 18 New Zealand rabbits were clamped to make model of sciatic nerve injury which is Sunderland III damage, and put them into the model group, the treadmill group and jiaji electroacupuncture combined with treadmill, while the sciatic nerve of 6 New Zealand rabbits were not clamped, just cut soft tissue around the sciatic nerve. Three days after modeling, the rabbits in treadmill group were treated by treadmill, The rabbits in jiaji electroacupuncture combined with treadmill group were treated by treadmill (the same to treadmill group) and jiaji electroacupuncture (electroacupuncture beside L5-L7 of the rabbits), whereas the rabbits in sham group and model group did nothing, but just feed.4weeks after treatment or feed, bone mineral density and bone strength of tibia were measured, Observe the tibia morphology slice and Immunohistochemical detection of neurotransmitter expression of SPResult1.Rabbit tibia bone strengthThere were significant reduction in bone strength of tibia in model group, treadmill group and jiaji electroacupuncture combined with treadmill group compared with sham group(P<0.05); and There were significant higher in bone strength of tibia in treadmill group and jiaji electroacupuncture combined with treadmill group compared with the model group(P<0.05); and there were significant higher in bone strength of tibia in jiaji electroacupuncture combined with treadmill group compared with treadmill group (P<0.05)2. Rabbit tibia bone mineral densityThere were significant reduction in bone density of tibia in model group, treadmill group and jiaji electroacupuncture combined with treadmill group compared with it in sham group (P<0.05); and There were significant higher in bone mineral density of tibia in treadmill group and jiaji electroacupuncture combined with treadmill group compared with the model group (P<0.05); and there were significant higher in bone mineral density of tibia in jiaji electroacupuncture combined with treadmill group compared with treadmill group (P<0.05)3.HE staining to observe morphology of tibia4 weeks after feeding, sham group suggest that the normal trabecular bone structure stability, the bone trabecular is thick and most of the trabecular bone connection points, cortical bone extension is far, structural stability is strong, supporting role is obvious. Model group suggest that bone trabecular small or thin shrink, The trabecular bone gap is bigger, less of the trabecular bone connection points, the worse of the trabecular bone structure stability.4 weeks after treatment, the results of treadmill group and jiaji electroacupuncture combined with treadmill group suggest that Trabecular bone biomechanical indexs are better than the model group, for example, The trabecular bone is thicker, The trabecular bone gap is smaller, more of the trabecular bone connection points, the better of the trabecular bone structure stability, but the indexs of the sham group are better than the two groups’;and the jiaji electroacupuncture combined with treadmill group is better than the treadmill group.4.Immunohistochemical detection of neurotransmitter expression of SP4 weeks after treatment, model group is less than the other three groups, the differences were statistically significant (P< 0.05), sham group is more than jiaji electroacupuncture combined with treadmill group and treadmill group, the differences were statistically significant (p<0.05), and there were significant higher in bone content of SP of tibia in jiaji electroacupuncture combined with treadmill group compared with treadmill group (p<0.05)Conclusion1.treadmill group and jiaji electroacupuncture combined with treadmill group all can promote the tibial bone morphology of tibia after peripheral nerve injury2. jiaji electroacupuncture combined with treadmill group can promote bone strength and bone mineral density of tibia of rabbit with sciatic nerve injury,and it is better than treadmill group in effectiveness.3.That jiaji electroacupuncture combined with treadmill group promotes sciatic nerve recovery and increases the expression of SP in the tibia of rabbit may be one of the mechanisms which improve the tibia bone mineral density and bone strength.
