Objective: To observe the effect of Jiaji electroacupuncture combined with neuroloening on the behavioral scores of rabbit sciatic nerve after compression injury,toluidine blue staining to detect the sciatic nerve axillary count and the expression of GAP-43 and MBP.The related mechanism of loosening technique on the regulation of axon regeneration after sciatic nerve injury in rabbits provides a theoretical basis for the treatment of sciatic nerve injury by Jiaji electroacupuncture combined with nerve loosening technique.Methods: A total of 180 New Zealand rabbits were randomly divided into5 groups by clamp method.Two groups in the control group: normal control group and model control group;treatment group 3 group: Jiaji electroacupuncture group The nerve relaxation group,the Jiaji electroacupuncture combined with the nerve relaxation group,36 in each group.According to the 1 week,2 weeks,and 4 weeks after surgery,they were further divided into 3 subgroups,12 in each group.The normal control group did not do any treatment,and the model control group was placed in a cage and kept quietly.The nerve relaxation group was treated with nerve mobilization,the electroacupuncture group was treated with electroacupuncture,and the electroacupuncture combined with nerve relaxation group was performed with Jiaji electroacupuncture and nerve mobilization.After 1 week,2 weeks,and 4 weeks of treatment,behavioral scores were obtained.Toluidine blue staining of sciatic nerve axons were counted,and GAP-43 and MBP protein expression was detected by immunohistochemistry.Results:(1)Toe reflex and modified Tarlov score: The scores of the treatment group were higher than the model group,which was lower than the normal control group(p<0.05).The Jiaji electroacupuncture combined with nerverelaxation group was higher than the single-spine electro-acupuncture group and single item.The nerve relaxation group(p<0.05),with the best recovery at 4 weeks.(2)Axon regeneration: the axon regeneration in the treatment group was better than that in the model group,the difference was statistically significant(p<0.05),and the electroacupuncture combined with nerve relaxation group was higher than the Jiaji electroacupuncture group and nerve mobilization.Groups(p < 0.05),of which 4 weeks time point regeneration was the best.(3)GAP-43 protein expression: Compared with the normal control group,the expression of GAP-43 protein in the other groups increased,the difference was statistically significant(p<0.05);compared with the model control group,the three treatment groups The expression of GAP-43 in the sciatic nerve was increased,the difference was statistically significant(p<0.05);and the expression of GAP-43 in the electroacupuncture combined with nerve relaxation group was better than that in the Jiaji electroacupuncture group and the nerve mobilization group.The difference was statistically significant(p < 0.05).(4)MBP protein expression: Compared with the normal group,the expression of MBP protein in the other groups decreased,the difference was statistically significant(p<0.05).Compared with the model control group,the expression of MBP in the sciatic nerve of the three treatment groups increased.The difference was statistically significant(p<0.05),and the expression of MBP in the electroacupuncture combined with nerve relaxation group was better than that in the Jiaji electroacupuncture group and the nerve mobilization group(p<0.05).In conclusion:(1)Jiaji electroacupuncture,nerve loosening technique,Jiaji electroacupuncture combined with nerve mobilization can improve thebehavioral score after sciatic nerve injury in rabbits,promote the regeneration of injured axons,and increase the growth-related factor GAP-43 protein and myelin sheath.Expression of the basic protein MBP.(2)Jiaji electroacupuncture combined with nerve mobilization can promote the repair of neurons by increasing the expression of growth factor GAP-43protein;promoting the regeneration of myelin by increasing the expression of myelin basic protein MBP,thereby promoting the damaged peripheral nerve Repair.
