The Research Of The Protection Of Zhishi Xiebai Guizhi Decoction Extract On Isolated Rabbit Cardiac Ischemia-Perfusion

Objective:Preparation of myocardial ischemia-reperfusion injury model use Langendorff solated heart perfusion technique,to observe the effects of different doses of Zhishi Xiebai Guizhi Decoction extract on isolated rabbit’s heart ischemia-reperfusion injury, and to explore the possible mechanism and provide experimental data for clinical safe, scientific, objective and reasonable use of Zhishi Xiebai Guizhi Decoction in the treatment of ischemic heart disease.Method:Detecting ECG Ⅱ of healthy adult rabbits by BL-420F organism function experiment system, to reject those objects whose ECG is abnormal, and those whose ECG is normal are random, establish rabbit Langendorff solated perfused cardiac model, divided 5 groups:(n= 10), normal group, model group, high dose group, normal dose group, low dose group.Normal group:perfuse isolated cardiac with Locke solution for 125min; Model group:perfuse isolated cardiac with Locke solution for 20min, stop for 15min, Locke solution perfuse for 90min; Low-dose drug group, dose group and high dose group:treatment method was the same as that of the model group, but the final step of reperfusion, added different doses of Zhishi Xiebai Guizhi Decoction extract in Locke’s solution, record before their stopping irrigation, stop irrigation and again during reperfusion for 1 min,10 min,20 min,30,40,50,60,70, 80,90 of electrocardiogram and ventricular pressure, coronary flow and myocardial water content percentage.Results:1.Rabbits’HR decrease significantly in the model group ECG, the P-R interval increase, ST increase distinctly, T wave range increase. It exist significant difference compared with normal control group. Rabbits’LVSP decreased significantly, LVEDP increase distinctly, ±dp/dtmax decrease, after statistics analysis there are a significant difference between the model group and the normal group.Water content of myocardial issue of model group apparently decrease, it exist significant difference compared with normal control group. Coronary blood flow of model group decrease apparently, after statistics analysis there is a significant difference between the model group and the normal group.2.Low dose group and normal dose group of Zhishi Xiebai Guizhi Decoction extract, In ECG HR accelerate, ST segment depression, T wave amplitude decreased, compared with the model group, it exist statistically significant difference; Low dose group and normal dose group of Zhishi Xiebai Guizhi Decoction extract, LVSP increased, LVEDP reduced,+dp/dtmax faster, compared with the model group there are significant differences by statistical analysis; Normal dose group of Zhishi Xiebai Guizhi Decoction extract -dp/dtmax faster, compared with the model group there are significant differences by statistical analysis;Low dose group and normal dose group of Zhishi XieBai Guizhi Decoction myocardial water content are decreased, compared with the normal group, there is significant difference by statistical analysis; Low dose group and normal dose group of Zhishi Xiebai Guizhi Decoction, coronary blood flow significantly increase, compared with the normal group, there is significant difference by statistical analysis.3. The change of indexes of high dose group some rabbits are similar to low dose group and normal dose group before 30 min. In High dose group some rabbits occurs arrhythmia, so there is no statistical analysis.Conclusion:1. We established the model of myocardial ischemia reperfusion injury in rabbits successfully. Performance of HR decrease, S-T segment and T wave raise, P-R interval prolong, Left ventricular function have changed, coronary blood flow is significantly decreased, the percentage of myocardial water content is increased.2. Low dose (2.5ml/kg) and normal dose (5ml/kg) of Zhishi Xiebai Guizhi Decoction extract have a protective effect on myocardial ischemia reperfusion model in rabbits, they can improve the myocardial contractility, improve heart pump function.
