Objective:Using Langendorff isolated heart irrigation flow technology, copy of myocardial hypoxia model and observation of different doses of Zhishi Xiebai Guizhi Decoction extract on rabbit isolated perfused heart in hypoxia, safe and rational clinical use of Fructus aurantii Immaturus scallion white Guizhi Decoction in the treatment of ischemic heart disease and provide scientific and objective experimental data.Methods:Firstly, the II lead was detected by BL-420F biological function test system, the ECG was removed and the Langendorff isolated perfused cardiac model was duplicated.Randomly divided into 5 groups (n=10):normal group, model group, high dose group, dose group, low dose group. Normal group:after leaving the heart perfused with 20min pulse stability, Locke’s fluid sustained reperfusion for 30min; model group:from the heart Locke’s solution perfusion 20 min after heart beating stable, stop oxygen perfusion 20min, Locke’s solution hypoxia and reperfusion for 30min; drugs with low dose group, middle dose group, high dose group:treatment method was the same as that of the model group but then perfusion with Locke’s solution were added different doses of Zhishi Xiebai Guizhi Decoction extract, were recorded respectively before hypoxia,20min hypoxia after 1min,5min, 10min,15min,20min,25min,30min ECG, left ventricular pressure, coronary flow and myocardial water content percentage.Results:1, The model rabbits ECG HR slowed down, S-T segment elevation, T wave amplitude elevation, P-R interval prolongation, compared with normal group there were significant differences were statistically; the model group decreased LVSP, LVEDP significantly increased presence of relatively slow down ±dp/dtmax, and the normal group significantly different; the model group decreased coronary blood flow, there is a significant difference in comparison with the normal group; the model of myocardial water content percentage increased compared with normal group there was a significant difference.2, The Zhishi Xiebai Guizhi Decoction extract of low dose group, middle dose group HR, shortened P-R interval, accelerate S-T segment depression, T wave amplitude decreased, compared with the model group, the statistical analysis of significant difference; Xie Bai Guizhi Decoction extract of Fructus aurantii Immaturus low dose group, middle dose group, LVEDP decreased,+dp/dtmax LVSP increased faster, faster-dp/dtmax, compared with model groups after statistical analysis there was a significant difference; Zhishi Xiebai Guizhi Decoction low dose group, middle dose group myocardial water content percentage decreased, compared with the model group, the statistical analysis of significant difference; Zhishi Xiebai Guizhi Decoction low dose group, middle dose group significantly increased coronary flow compared with the model group, there were significant differences by statistical analysis.3,The Zhishi Xiebai Guizhi Decoction extraction solution in high dosage group and hypoxia after 5min,10 min time point and the model group compared with the significant difference,15min after only rabbits showed arrhythmia, hence not do statistics.Conclusion:1, The present study with hypoxic perfused rabbit model,it performance of slowing HR, between S-Tsegment elevating, T wave height tipping,P-R prolongating.Left ventricular function significantly changed, Significantly decreased coronary flow, percentage increase in myocardial water content.2,The Zhishi Xiebai Guizhi Decoction Extract low-dose group (0.395g/kg) and normal dose group(0.79g/kg) have a protective effect for Hypoxic perfused rabbit model and they can promote heart function recovery.3,The Zhishi Xiebai Guizhi Decoction Extract high-dose group(1.58g/kg) has a protective effect for Hypoxic perfused rabbit model but it can cause arrhythmia.
