| BackgroundAcute lung injury (ALI) is a clinical common acute critically ill, its pathology characteristic is too much out of control inflammatory response in lungs and Pulmonary capillaries permeability increased, involving in inflammatory cells’migration, gather and the release of inflammatory mediators and so on.There are respiratory distress and progressive hypoxemia in clinically, Severe cases can rapidly develop into acute respiratory distress syndrome and even life threatening. Tumor necrosis factor-a(TNF-a) and intercellular adhesion molecule-1(ICAM-1) play an important role in development in ALI. Curcumin is the active ingredient of the traditional Chinese medicine curcuma, A large number of clinical trials to prove its scavenging free radicals, anti-oxidation, anti-inflammatory, antitumor, anti fibrosis, and can reduce all kinds of synthesis and secretion of inflammatory mediators such as many pharmacological functions. explore curcumin treatment of molecular mechanism of acute lung injury, for the clinical application of curcumin to provide a theoretical basis.ObjectivesTo investigate the protective effect of curcumin on SD rats with acute lung injury(ALI) induced by bleomycin(BLM),and the influence of curcumin on the expressions of two of inflammatory factor of TNF-a and intercellular adhesion ICAM-1.Methods1. Molding and drawing materials:The Experiment selected SD rat of 140~160g healthy male as the research object.The SD rats were randomly divides into sham operation group,model group,curcumin high, medium and low dose group(12 in each group). Apart from the sham group of SD rats given 1-time intratracheal infusion of equal volume of saline,all other group SD rats were given 1-time intratracheal infusion of BLM.After 24 hour times, we started to administered that curcumin high-, medium-, low-dose group were dosed with curcumin at doses of 200,100,50 mg/(kg-d),stopping until the model set up to the day the animals were sacrificed.and the same time,the sham operation group and model group were fed with an equal volume of saline.Until at 3rd and 7th day,half of SD rat of all groups were respectively killed.2. The Pathological changes of SD rat’s the lung tissue were observed by HE staining. The two kinds of protein expression situation about TNF-a and ICAM-1 in SD rats lung tissue was detected by immunohistochemistry.3. The expressions of TNF-a and ICAM-1 in SD rats lung tissues were analyzed by Western blot.Results1. Compared with the sham group, Model group rats in behavioral performance for depression, unresponsive, shortness of breath, hair erect, Feeding reduced. On the change of lung tissue morphology, gas exchange function of model group rats lung tissue seriouly declined, most of the consolidation of the lung, alveolar show the lung interval obvious thickening and gas reduced.Model group expressed pulmonary congestion, edema of tissue, inflammatory cells (mainly neutrophils and lymphocytes) infiltrating and significant inflammation.2. Compared with model group,To observed treatment effect of three curcumin group were by HE staining, The damage degree of curcumin high, medium and low dose group had different degrees of improvement, Treatment effect of curcumin medium dose group was better than that of curcumin high and low dose group.3. To observing protein expression level of TNF-a and ICAM-1 in each group of SD rats lung tissue were detected by immunohistochemistry and Western blotting test methods:Protein expression of TNF-α 和 ICAM-1 level in SD rats model group were significantly higher than that of sham operation group. Curcumin high, medium and low dose can obviously reduce the expression of two proteins. The difference is statistically significant (p<0.05). The two kinds of protein expression level in Curcumin contrast between high, medium and low group:medium dose group< high dose group< low dose group, medium dose group effect is the most obviousConclusions1. Curcumin is a preventive protection function in the process of the development of ALI.2. its mechanism about protection function of Curcumin may be related to inhibiting inflammatory cytokines of the expression of ICAM-1 and TNF-a. |
