Background:Recently, with the economic development and the change of people’s lifestyle and eating, acute myocardial infarction incidence in China increased year by year, becoming an important killer of human life. With the rise of biomedicine, bone marrow mesenchymal stem cells had been used to cure myocardial infarction for decades, in the past, researchers just focused on the therapeutic effect of BMSCs mixed groups, paying little attention to some subpopulation.The relationship and treatment mechanism between subpopulations and mixed group need to be further studied.Objective To explore the therapeutic effect of CD+29 bone marrow mesenchymal stem cells on pigs with myocardial infarction, to provide the basis for the clinical application of BMSCsMethods:1. BMSCs were isolated by density gradient centrifugation and cultured in vitro, passaged to the third generation, using flow cytometry to select the subpopulation of CD29+BMSCs then cultured continually.2.12 Bama pigs were randomly divided into CD29+BMSCs group and BMSCs group. Acute myocardial infarction(AMI) models were established by the method of blocking the left anterior descending coronary artery with cardiac catheter interventional balloon. CD29+BMSCs/BMSCs were injected directly into the infarct area. To observe capillary density and angiogenesis of infarct area. Cardiac function was detected before and 4weeks after operation.3. Observed the pathological change of infarction area by HE and Masson staining. The change of Ⅷ factor at the infarction edge in pigs heart was detected by immunohistochemistry.Results:BMSCs highly expressed CD29, hardly expressed CD34.4 weeks after operation the cardiac function of CD29+BMSCs group was better than that of BMSCs group, significant difference (P< 0.05). the second and third weeks, the amount of VEGF expression was highest, compared with BMSCs group, the difference was statistically significant (P< 0.05), the forth week, the VEGF decreased to the level of before operation, no obvious difference in the two groups(P> 0.05). CD29+ BMSCs group expressed Ⅷ factor higher than that of BMSCs, significant difference (P< 0.05).Conclusions:Compared with BMSCs group, CD29+ BMSCs subpopulation can more obviously promote angiogenesis of the infarction area, increase the number of new blood vessels, improve cardiac function. the treatment effect is better than that of BMSCs mixed group...
