BackgroundCandida albicans (C.albicans) is the most common clinical fungal infections, mannan-binding lectin (MBL) is a plasma protein secreted by liver cells, which belongs to the C type lectin superfamily collectin family, is a natural one key of soluble PRR in the immune system, known as "the key component of the innate immune system". Regulatory T cells (Tregs) and its phenotype was CD4+CD25" Foxp3+, accounted for 5%~15% of the CD4+ T cells in total, is the cell subsets of CD4+T, which has immune regulation function in the human body. Our prophase research proof that, MBL can modulate the C.albicans phase transformation and its immune response. There need further research to make clear that how plasma concentration of MBL changed in the patients with C.albicans, how express of Treg cells changed, and whether there is a correlation between this two factors. The study will reveal the role of MBL in regulating immune response against C.albicans infection.In this study, peripheral blood samples on clinical C.albicans infection patients and healthy controls were collected firstly, with further detection of MBL concentration in plasma by ELISA. Then, analysis the expression of Treg in PBMC cells by flow cytometry, RT-PCR analysis the expression of Treg cell specific transcription factor Foxp3, finally uses ELISA to detect the expression levels of Treg cells inflammatory factor IL-1 beta. The results convinced that, compared with the healthy control group, plasma concentration of MBL in patients with C.albicans infection was significantly increased, and the expression of Treg cells in C.albicans infected patients in PBMC was significantly increased, the expression of Treg cell specific transcription factor Foxp3 are increased, Treg cell related inflammatory factor IL-1 beta secretion are also increased. The results show that, in the case of C.albicans infection, when MBL elevated, the expression of Treg cells elevated. Which suggested that, in the case of C.albicans infection, there was positive correlation between the changes of MBL concentration and the expression levels of Treg cells. The study provides new theoretical basis and a new target for clinical treatment of C.albicans infection.ObjectiveAnalysis the change of MBL concentration and the expression of Treg cells in plasma of patients with C.albicans infectionMethods1. Collecting clinical specimens and detecting the concentration of MBL:collect 20 cases of peripheral blood samples in both C.albicans infected patients and healthy controls according to the inclusion criteria. Then detect the level of MBL in peripheral blood by ELISA.2. Analysis Treg cells by flow cytometry:collect every blood sample, adding FITC-anti CD4 and PDC- anti CD25 and PE- anti Foxp3 monoclonal antibody into this samples, then analysis the expression of Treg cells by flow cytometry.3. Detect levels of IL-1 beta 3 by ELISA:detect the levels of IL-1 beta 3 by ELISA patients and healthy controls, the level of peripheral blood inflammatory cytokines IL-1 beta ELISA for detection of C.albicans infection.4. Analysis the expression of Foxp3 by RT-PCR:analysis the expression of Treg cell specific transcription factor Foxp3 of both C.albicans infected group and healthy control group by RT-PCR.5. Statistical analysis:statistical analysis according to the data (x±s). Use SPSS 19 to carry on the analysis, and use One-way ANOVA test to compare the multiple samples, with P< 0.05 for the difference has statistical significance.Results1. the average MBL serum concentration of 20 patients was 1.893 μg/ml, the average MBL concentration of 20 cases of the control group was 1.310 μg/ml, and there were statistical significance compared (P<0.05). The plasma concentration of MBL in patients with C.albicans infection displaying significantly increased.2. Compared with the healthy control group, the percentage of CD4+CD25+Foxp3+ cells significantly increased (P<0.05) in the Peripheral blood PBMC 2 of C.albicans infected patients, which suggested that the numbers of Treg cells in patients with C.albicans infection in PBMC ratio increased.3. Compared with healthy control group, Treg cell inflammatory factor IL-1 beta levels in the plasma of patients with C.albicans infection increased significantly, the difference was statistically significant (P<0.05), suggesting that C.albicans infection of Treg cell reactivity enhanced.4. the Foxp3 expression of C.albicans infected patients is higher than that in healthy control group (P<0.05), suggesting that Treg cell specific transcription factor Foxp3 expression enhanced in the PBMC of C.albicans infected patients.ConclusionIn the case of C.albicans infection, the concentration of MBL was significantly increased, the expression of Treg cells was also increased.
