Significance Of Expression Of Emt Related Protein In Laryngeal Carcinoma Under Hypoxia

[Objective] The main objective of this paper is to investigate the effect of the hypoxia on EMT and the clinical significance of expression of the related protein in laryngeal carcinoma.[Methods] After laryngeal carcinoma SCC10 A cell culture had been under hypoxia, the invasion and migration ability of SCC10 A cell were detected by Transwell assay. The expression of HIF-1α and EMT protein(N-cad, Vim, E-cad) were measured by Western blot. The immunohistochemical technique was used to examine the expression of GLUT-1, Vim, E-cad and N-cad in the normal larynx squamous, laryngeal squamous cell carcinoma and pericarcinous tissue, then to explore the relationship between the expression of these protein and the clinicopathological features of laryngocarcinoma patients.[Results] After SCC10 A cells were treated under hypoxia for 24 h, the results of Western blot showed that the expression of HIF1α, Vimentin and N-Cad increased significantly(P<0.05),compared with normoxia group.And the ability of invasion and migration was significantly promoted(P<0.05). In 57 tumor samples,GLUT-1 expression of 49 patients(85.96%) was observed. However, there were respectively 4 cases(8%) and 12 cases(21.05 %) in normal tissue and pericarcinous tissue. The position rate of GLUT-1 in the three groups were significant different(P<0.05), and further analysis found that GLUT-1 expression was statistically significant correlated with the clinical stages of laryngeal cancer and lymph node metastasis(r=0.424, P<0.05; r=0.340, P<0.05). The expression of Vim and N-cad was weakly positive in normal laryngeal squamous cell epithelium in 5 cases(10%). E-cad intense positive expression was in the 50 cases(100%). The expression of Vim, N-cad and E-cad was similar in normal tissue and pericarcinous tissue(26.32%; 29.82%; 82.46%). Vim and N-cad expression was strongly positive(82.45%; 84.21%). And E-cad expression has weakly positive(21.05%). Vim had a significantly positive correlation with the expression in clinical stages of laryngeal cancer and lymph node metastasis(r=0.504, P<0.05; r=0.413, P<0.05), N-cad had simila result(r=0.376, P<0.05; r=0.467, P<0.05). But E-cad was significantly negatively correlated with the expression in laryngeal squamous cell carcinoma( r=-0.319, P<0.05). Patients with positive GLUT-1,Vim and N-cad expression had significantly lower survival rate than those with the negative expression(P<0.05), and patients with positive E-cad expression had higher survival rate than those with negative expression(P<0.05). The results showed that there was a significant positive correlation between the expression of GLUT-1 and Vim and N-cad in laryngeal squamous cell carcinoma(r=0.600, P<0.05; r=0.379, P<0.05). GLUT-1 had no relation to E-cad in laryngeal squamous cell carcinoma(P>0.05).[Conclusions] Hypoxia promotes invasion and metastasis of laryngeal carcinoma cells, and induces EMT in laryngeal carcinoma cells. GLUT-1, Vim and N-cad has a significantly positive correlation with clinical stages of laryngeal cancer and lymph node metastasis in laryngeal squamous cell carcinoma. But E-cad is significantly negatively correlated with the expression of Glut-1 in laryngeal squamous cell carcinoma. GLUT-1 has a significantly positive correlation with Vim and N-cad expression in laryngeal squamous cell carcinoma, while GLUT-1 has no correlation with E-cad expression in laryngeal squamous cell carcinoma. The survival rate is low in patients with positive GLUT1, Vim and N-cad expression in the laryngeal carcinoma; the survival rate is high in patients with positive E-cad expression.
