Integration of multiple imaging modalities was an efficient way to achieve precise and fast imaging targets in biomedical diagnosis. In recent years, many multimodal imaging agents had been developed and exhibited superb imaging capabilities. Among the various combinations of imaging modalities, the combination of 19 F magnetic resonance imaging(19F MRI) and fluorescence imaging was highly advantageous. In this paper, we developed a series of fluorinated amphiphiles as 19 F magnetic resonance /fluorescence dual imaging agents and detected their in vitro 19 F MRI properties, optical properties, sizes in water and biological toxicity.Three 19 F magnetic resonance / fluorescence dual modal imaging agents were synthesized(agent A1, A2 and A3). The results indicated that A1 possessed the weakest 19 F MRI sensitivity due to strong self-assembly. After structural optimization A2 and A3 exhibited better 19 F magnetic resonance imaging sensitivity due to different self-assembly modes. A2 had weaker UV absorption and fluorescent intensity. A1 and A3 had the similar UV absorption, but A1 showed stronger fluorescent intensity. Dynamic light scattering indicated the sizes of agents A1, A2 and A3 were 307.6 nm, 95.1 nm and 70.9 nm. A3 showed no acute toxicity on Hela cells and mice.We synthesized a promising imaging agent A3 with high aqueous solubility, high 19 F MRI sensitivity, preferred fluorescent properties and low toxicity, which might have broad applications.
