Effect Of “Warming Yang And Nourishing Qi” Acupuncture To EAMG Rat Neuromuscular Junction MRNA Expression Of Acetylcholine Receptor

Purpose: acupuncture is a promising method to cure the autoimmune myasthenia gravis(MG). In my thesis, we will give a scientific evaluation of the acupuncture to cure the MG. We would treat the experimental autoimmune myasthenia gravis(EAMG) rat with “warming yang and nourishing qi” acupuncture method combining the Real-time PCR techniques to test and analyze the mRNA of EAMG rat neuromuscular junction(NMJ) acetylcholine receptor. Furthermore, we will discuss the dynamics of how the “warming yang and nourishing qi” acupuncture affects the EAMG; and then, we will provide an enhanced rule to solve the MG disease in clinic, and an effective criterion for the clinical diagnosis and improving judgment.Method: We will construct 60 EAMG SPF- “grade three” femaleLewis rats by activating them with AchRα1 129-145 polypeptide fragments. Select 30 EAMG rats randomly and divide them into 3 groups, labeled as model group, drug group and acupuncture-treating group, respectively. Another 10 SPF-“grade three” female Lewis rats, which are raised together with EAMG rats, are used as the comparing group. In the experiments, there is no treatment to the comparing group and the model group. Acupuncture-treating group will be treated with “warming yang and nourishing qi” acupuncture method. And pyridostigmine bromide is used to cure the drug group rats by gavage. After two courses of treatment, we will take the neuromuscular junction of the rats as testing material, to detect the acetylcholine receptor mRNA with Real-time PCR techniques and analyze the difference among groups quantitatively.Result: ①. In the constructing step, we succeed to obtain 32 EAMG rats with the successful rate of 53.3%; ②. The Real- time PCR results show us that the γ- and ε-acetylcholine receptor mRNA from both of model group and acupuncture-treating group have significant increasing(P<0.01); ③. The γ- and ε- acetylcholine receptor mRN A from both of drug group and acupuncture-treating group have significant increasing(P<0.01).Conclusion: ①. The amount of acetylcholine receptor mRNA from the neuromuscular junction is closely linked to EAMG condition. ②. “warming yang and nourishing qi” acupuncture therapy could effectively improve the level of acetylcholine receptor mRNA. ③.“warming yang and nourishing qi” acupuncture therapy is better than pyridostigmine bromide drug treatment to affect the acetylcholine receptor mRNA level of EAMG rat.
