In the present study, experimental autoimmune myasthenia gravis (EAMG) was induced. In order to discuss the effect of polyclonal antibodies to BLyS on prevention of EAMG and on treatment of ongoing EAMG, polyclonal antibodies to BLyS were prepared and used in EAMG prevention and treatment experiments. The mechanism of polyclonal antibody to BLyS in EAMG was studied initially. The results were summarized as follows:1 Induction and evaluation of experimental autoimmune myasthenia gravis (EAMG)C57BL/6 mice were immunized with peptides of α-subunit of acetylcholine receptor and naked bacteria. At 14th day after the second immunity, the clinical manifestations were observed. The titers of antibody of acetylcholine receptor, swimming time and electromyogram (EMG) were detected. After the second immunity, most of the model group mice were a little easy to fatigue and the level was1 mark. The titers of antibody of 12 mice in 20 was positive and the difference was significant compared with control group. During the swimming experiment 18 model group mice in 20 were to be positive. All of the EMG of model group mice were to be positive and the greatest percent decrement was 82%. It was successful to induce EAMG and prepared to discuss the mechanism of EAMG.2 Preparation and purification of polyclonal antibodies to BLyS and to BSAJapanese big ear white male rabbits (2~3 kg) were immunized from lymph nodes or subcutaneous parts. Antiserum were prepared, the titer was detected by indirect ELISA. Antibodies were purified with saturated ammonoium sulfate. Polyclonal antibody to BLyS was obtained and the antisera titer reached to 1 : 5 × 105; polyclonal antibody to BSA was obtained and the antisera titer reached to 1 : 5× 10~4. Polyclonal antibodies to BLyS and to BSA were prepared successfully and were prepared to discuss the correlation between BLyS and EAMG.
