Effect Of Yiqi Jiedu Decoction Containing Serum On Proliferation Of Mouse Hepatocellular Carcinoma H22 Cells In Vitro

Purpose: Modern TCM clinical treatment of multiple according to the theory of TCM by Qi and Chinese medicine decoction of clearing away heat and toxic material,promoting blood circulation Sanjie class of traditoonal Chinese medicine in the treatment of liver cancer.In this experiment,using the method of serum pharmacology to observe the effect of iqijiedu in vitro,and to explore the mechanism of anti Yiqijiedu in the treatment of hepatocellular carcinoma,and open up a new path for the Chinese medicine treatment of liver cancer.Method: Take health 10 rats, 4 consecutive days to 1.25g/ml crude drug concentration wiil YiqiJiedu Decoction extraction liquid at a dose of 10ml/kg intragastric administration.Onthe last administration after 1 hour to 0.9% sodirm pentobarbital sodium pentobarbital intraperitoneal injection and 80 DEG C to preserve the reserve;by the same methods continuous 4 Tiandeng dose of mormal saline by gavage prepared negative control serum.H22 cells were divided into 5 groups:the negative control group,the positive control group(5-Fu),the Chinese medicine high,middle and low dose group.① By inverted microscope observation of each group containing serum on cultured H22 Cell Growth and morphology;② The effect by MTT method,CCK-8 method were used to detect the serum on H22 cell proliferation;③ Through the influence of cycle inhibitory serum was detected by flow cytometry on the growth of H22 cells;④ The RT-PCR method of medicated serum on in vitro culture of mouse hepatoma H22 Cell proliferation inhibition of apoptosis related proteins.Result:Yiqi Jiedu Decoction medicated serum on in vitro culture of mouse hepatoma H22 cells proliferation in different concentrations have certain inhibition.① The morphology and growth state of H22 cells of mice liver cancer cells in vitro were inhibited;② The proliferation of H22 cells in vitro was inhibited and the inhibition rate wasincreased;③ The inhibition of H22 cell growth cycle in mice was mainly inhibited by the inhibition of G0/G1 phase;④ It has a certain effect on the apoptosis of mouse hepatoma H22 cell related pretein in vitro,down regulate the expression of Bcl-2, Bax expression.Conclusion: The experiment shows Yiqijiedu cultured mouse hepatoma H22 cells can inhibit the proliferation of in vitro.The possible mechanism of Yiqijiedu Decoction drug containing serum cultured mice hepatocellular carcinoma cells H22 in vitro through inhibitiion of G0,G1 and H22 og cell division and apoptosis related protein Bcl-2 and Bax.
