| Objective:To establish periphery-central inflammation network and to illuminate the therapeutic effect of Yiqijiedu Formula to ischemic stroke basing on mRNA and miRNA technology.Methods:1.Basing on the analysis of data,network signaling pathways of mRNA,including whole gene networks,inflammation related networks and organ-related networks were established.The steps are as follow.Setting fold change value as 1.3,then the satisfactory genes was selected for further study.These selected genes were in-orderly put into process of analysis:difference analysis,shared-parts analysis and richness analysis.And according to these analyses,networks were set up as background for next analysis.2.PCR was used to confirm the veracity of these mRNA networks and key hub-genes.3.Then put genes which express significantly in Yiqijiedu treatment group into the networks we have established in last steps,and compose the therapeutic regulation mRNA gene network of Yiqijiedu Formula for ischemic stroke.4.In addition,analyse the data of miRNA chips with same methods as step 1,and estimate the target mRNA genes of the miRNA which was up-regulated or down-regulated significantly.Then connect the targeted results with Yiqijiedu therapeutic regulation network,lead to a new network containing both mRNAs and miRNAs.Results and Conclusion:Firstly,our results spggest that there exists a network which is named as central-periphery inflammation network,and it may relate with inflammation basing on mRNA level and miRNA level regulation.This network has been established and verified by both mRNA chips analysis as well as PCR test.Secondly,we found that four pathways,including NF-kappa B signaling pathway、PI3K-Akt signaling pathway、Chemokine signaling pathway、Jak-STAT signaling pathway,were responsible for the regulation effect of Yiqijiedu Formula during ischemic stroke process.At the same time,a series of crucial inflammation-related genes were in miRNA network,which are the key ones regulated by miRNAs in stroke. |
