Objective: Rumex japonicus Houtt is one of most important traditional medicines of China, it is a medicinal materials resources also. Rumex japonicus Houtt is mainly used to anticancer, clearing away heat, toxic materials, anti-inflammation and others applications in clinical. It has a long history of clinical application in China. Recently research has shown that it has good antifungal, antioxidant,anticancer and anti-aging activities. But, there are not have so many research focus on the basis research such as chemical compounds or contents analysis and bioactivities of Rumex japonicus Houtt. In the recent years, with the modern extraction and separation technologies, spectroscopy technology and the rapid development of modern pharmacology, more and more new methods and modern technologies are application to the research of modern Chinese medicine. The chemical constituents and pharmacological activities of Rumex japonicus Houtt research will becoming more and more deeply and clearly. In order to better and more reasonable development and utilization of the traditional Chinese medicine(TCM), in this study, one of traditional Chinese medicine that Rumex japonicus Houtt was used as a material, we used that for extraction,separation compounds and evaluation of the antioxidant free radical scavenging activity,antioxidant activity,anti-inflammation activity and anticancer activity of the fractions of Rumex japonicus Houtt. And analysis the main effective components of each fractions.Method: In this study,we took dry Rumex japonicus Houtt crude drugs, adopt the traditional extraction method of traditional Chinese medicine(TCM) that water decoction extraction mode. Sample was mixture with water extraction for 1 hour, then filtered and got the water extract. Repeat this procedure threetimes and combine all of the water extracts. Concentration the water extract to a smaller volume and treatment with ethanol and keeping quiet place over night for precipitation. Filter the extracts and concentration it and get the dry extract. The extract was dispersion in water, and fraction by n-hexane and chloroform saturation by water, ethyl acetate saturation by water and n-butyl ethanol fract respectively. Then get fractions and concentration each fractions. Do the preliminary component analysis and biological activities scavenging of each fraction. According to the results of the chemical compositions of each fraction and biological activities analysis, we selecting better bioactive fractions as the target fractions. Using those target fractions for the further research such as separation and purification compounds and biological activities evaluation. In this study we are used thin layer chromatography(TLC), silica gel column chromatography to do further separation and purification and detection of target fractions. The compounds of the targets fractions were identification the chemical structure by NMR spectroscopy method. The bioactivities such as free radical scavenging activity,antioxidant activity, anticancer activity and anti-inframmatory activity of the target fractions were evaluation by chemical analysis and cell line experiment in vitro.Results: The aqueous extract from Rumex japonicus Houtt has a certain ability to scavenging free radicals, chloroform fraction and n-butyl ethanol fraction were shown a better scavenging free radicals and anti-inflammation and anti-tumor activity but without toxic or side effects on normal cell lines. Three monomer compounds has been isolated and identified from these two fractions. They are chrysophanol,(compound-1), physcion(compound-2) and rutin(compound-3) respectively. According to this results, it can be thought that Rumex japonicus Houtt has a good scavenging free radicals,antioxidant,anti-inflammatory, antitumor activities. And the toxic substances of Rumex japonicus Houtt can be removed by the polarity gradient solvent fraction. Chrysophanol, physcion and rutin are the main bioactive compound of chloroform fraction and n-butyl ethanol fraction. And the chloroform fraction and n-butyl ethanol fraction has shown a good scavenging free radicals and anti-inflammatory, antitumor activities and highly safety.
