| Objective:Through the application of high performance liquid chromatography (HPLC) to detect 12 kinds of major chemical compositions of chrysanthemums respectively, Ultraviolet and visible spectrophotometry (UV-vis) to detect total flavonoids of chrysanthemums, and atomic absorption spectrophotometry (AAS) to detect the heavy metals content of chrysanthemums, to complete the study of the chemical constituents of chrysanthemum, provide more information for quality evaluation of chrysanthemum herbs, and security guarantees for its clinical application.Method:With the method of HPLC to detect the content of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, luteoloside, isochlorogenic acid B, isochlorogenic acid A, apigenin-7-O-p-D-glucoside, isochlorogenic acid C, luteolin, apigenin, diosmetin, acacetin, and use methodology to study. Using UV-vis to detect total flavonoids of chrysanthemums, AAS to detect the content of Pb, As, Hb of chrysanthemums.Results:(1) Selecting acetonitrile-water-phosphate gradient system as the HPLC chromatographic conditions; velocity of flow: 1.0mL/min; detection wavelength was PDA timing wavelength (0.00~15min,327.0nm;15.00~30min,348.0nm;30.00~65min,327.0nm; 65.00~100min,340.Onm);column temperature as the room temperature.12 kinds of chemical compositions show a good linear relationship and the average recovery complies with the provisions. The determination method is simple, reproducible, and can be used for quantitative control of chrysanthemum. There are some differences of chemical composition between different habitats and species of chrysanthemum. In 14 kinds of chrysanthemum samples, Xin’an Dayang chrysanthemum have the highest content of chlorogenic acid, pearl chrysanthemumthe have the highest content of luteoloside, Gong chrysanthemum have the highest content of isochlorogenic acid A, Xin’an Xiaoyang chrysanthemum have the highest content of apigenin, Kaifeng Dayang chrysanthemum have the highest content of isochlorogenic acid C, Huai chrysanthemum have the highest content of luteolin, Luanchuan Hangbai chrysanthemum 3 have the highest content of apigenin, Huai chrysanthemum have the highest content of diosmetin, Luanchuan Hangbai chrysanthemum 2 have the highest content of acacetin. (2) The method to measure total flavonoids of chrysanthemums is A1(NO3)3 coloring, at UV-vis 510nm. The results show that flavonoids of chrysanthemum vary from different origins and different varieties, the highest is Kaifeng Dayang chrysanthemum(5.71%), the lowest is Luanchuan Hangbai chrysanthemum(1.17%), and the Huai chrysanthemum is 2.48%. (3)Using the method of GF-AAS to detect Pb, and hydride generation-atomic absorption spectrophotometry HG-AAS to detect As, Hg. The results show that the content of Pb and As of 14 kinds of chrysanthemum samples meet the national green standards and Xin’an Dayang chrysanthemum, Xin’an Yejin chrysanthemum, farm Gong chrysanthemum, Kaifeng Dayang chrysanthemum content excessive Hg than the standards.Conclusion:The application method for the determination in this paper is accurate, reliable, and reproducible. In favor of complete the study of the chemical constituents of chrysanthemum, enhance and enrich more information for quality evaluation of chrysanthemum herbs, provide more comprehensive quality scientific evaluation of chrysanthemums, and reliable reference for quality assessment of chrysanthemum. |
