In this study, we prepared Gynura divaricata powder using freeze dryer. Then, the crude polysaccharide content(26.69+0.53mg/g) and total flavonoids(33.59+2.64mg/g) of the powder were determined by Phenol-sulfuric Acid Method and Na NO2-Al(NO3)3-Na OH solution respectively. The high fat and sugar diet plus STZ-induced were made diabetic mice. Subsequently, the mice were randomly divided into five groups: normal control group, diabetic model group, the powder at doses of 1.2%, 2.4% and 4.8% groups. They were given normal diet and normal water daily except the difference doses of powder group. Finally, blood was collected from the portal vein of eye and then pulled the neck to death.Their livers, pancreases, spleen were excised and weighed. The results confirmed that made diabetic mice method have lower toxicity and animal survival rate is higher. The powder can effectively reduce blood glucose and increase GSH-PX, SOD level in liver, at the same time, the content of MDA is decreased. In addition, it can reduce the resistance of insulin and regulate hepatic glycogen metabolism in diabetic mice. From the perspective of molecular biology, the expression of AKT, PI3 K and PDK-1 gene increased significantly. The protein expression of AKT, P-AKT, PI3 K and PDK-1 is consistent with m RNA. Therefore, we can speculate the hypoglycemic effects of Gynura divaricata by improving insulin resistance, enhancing the antioxidant ability and activating AKT/PI3 K signal pathway in diabetes mice.Therefore, the results proved the hypoglycemic function and mechanism of Gynura divaricata, this herb provide important scientific basis of diabetic mice development.
