| Object This research was designed to explore the better method to set up rabbit tumor model through abdominal incision using VX-2 tumor cells; to study the procedure of TACE for rabbits tumor model; to evaluate the safety and efficacy of TACE combined with Sorafenib powder in the treatment of the rabbits tumor model and to provide a theoretical basis for the treatment of hepatocellular carcinoma using sorafenib powder combined with TACE.Methods Tumor cells were delivered by microsurgical forceps and syringes separately to set up rabbit tumor model. Through comparing the advantage and feasibility of the two methods, the microsurgical forceps method was finally selected as the standard model building method in the formal experiment. There were total 32 rabbits and they were divided into 4 groups randomly, with 8 rabbits in each group.4 groups were respectively treated with iodized oil alone (TACE-i), iodized oil and pharmorubicin (TACE-ip), iodized and cis-DDP (TACE-ic), and iodized oil and sorafenib (TACE-is). Liver function test, kidney function test and CT scan were performed 3 days before and three days, one week, two weeks, and three weeks after TACE. The animals were sacrificed 3 weeks after TACE and pathological and immunohistochemical examination were performed. Comparison of the liver and kidney function before and after the treatment among the four groups was conducted to evaluate the security of sorafenib in TACE. And comparison of tumor size, VEGF, and MVD (CD34) among the four groups each before and after treatment was conducted to evaluate the efficacy of sorafenib in TACE-is.Results The damage of liver and kidney function in TACE-is group was not the most serious in four groups. The tumor volume was smaller 1 week after the operation (P<0.05) and did not increase significantly 3 weeks after the operation, while volume in other groups was obviously progressive (P<0.05). MVD (CD34) test results showed that TACE-is inhibited tumor angiogenesis (P<0.05), but the effect of inhibition of the expression of VEGF was not clear (P>0.05)Conclusion In this study, by comparing the microsurgical forceps method and syringe method to build rabbit VX-2 tumor model, microsurgical forceps method shows more advantages. The experimental skills of treating rabbit liver cancer by interventional therapy are established. By comparing the difference of liver function, kidney function, tumor size, VEGF and MVD (CD34) in TACE combined with sorafenib powder group and TACE with conventional TACE treatment group, it is concluded that the former is safer and more effective. |
