Objective: To study whether pregnant rats exposure to propofol cause offspring changes in space cognitive abilities and exploration abilities and phosphorylated tau Protein and beta-amyloid protein expression in hippocampal.Methods : 3-month Sprague-Dawley female rats(n=24)and male rats(n=8),which weight were over 200 g, were mated at same cage by the proportion of 3:1.Pregnant rats were randomly diveded into early group(group E),media group(group M),late group(group L) and control group(group C),with 6 rats in each group.Group E,M and L received propofol 80 mg/kg·d for 7 consecutive days.While in group C,propofol was replaced with equal volume physiological saline.Then the30-day offspring rats’ learning and memory were assessed used Morris water maze test. After that, offspring rats were sacrificed to determine the expression of phospho-tau(pSer202) and beta-amyloid(1-42) in the hippocampus by elisa and immunohistochemistry.Results: The lerning and memory abilities were declined significantly between group M and group E,group L, and group C(P < 0.05). Compared with group C,therewas difference in group E and group L on learning and memory abilities(P < 0.05). The result of immunohistochemistry : The beta-amyloid protein and phosphorylated tau hippocampal of group M ’offspring is significantly greater than that in other groups by elisa and immunohistochemistry(P < 0.05). Conclusions: Multiple propofol injections may impair rat offspring’ space cognitive abilities and exploration abilities, especiouly worse in mid-point of pregnancy, which may associated with increased beta-amyloid protein and phosphorylated tau protein.
