Objective:In the medical pure titanium and rabbit artificial joint prosthesis surface deposition of hydroxyapatite nano silver composite coating; sustained release performance observation of silver ion in the simulated body fluid; evaluation of composite tissue compatibility; to investigate the antibacterial properties of composite coating material of animal body. Methods:The use of pulse electrochemical method for preparing materials, to characterize the morphology, composition, space conformation.Materials will be immersed in simulated body fluid, energy absorption spectrometry to detect the concentration of Ag+ in different time points by atomic.According to national standards and ISO detection, tissue biocompatibility. In vitro for Staphylococcus aureus antimicrobial analysis. Adult New Zealand 48rabbits are randomly divided into experiment A, B, C in the three groups(n=16). For rabbit TKA operation, composite experiment A, B group implanted with different Ag+concentration of electrolyte deposition, experimental group C into the common artificial joint materials. To establish the model of rabbit TKA after operation of Staphylococcus aureus infection, antimicrobial property evaluation of composite animal in vivo..Result:0.5HA/Ag composite coating Ag content is 2.75wt%; first dayAg+ release accounted for 30d of total 45.55%; hemolytic rate is 1.64%, the in vitro antibacterial rate of Staphylococcus aureus is 95.3%. 1.0HA/Ag composite coating Ag content is 8.83 wt%; first dayAg+ release accounted for 30d of total 48.10%; hemolysis rate is 2.24%; in vitro antibacterial rate of Staphylococcus aureus is 97.6%. Animal infection models, experimental A, B group, CRP value, temperature and blood ESR value, PCT values are lower than the experimental group C, the difference is statistically significant (P<0.05). The rate of infection:experimental group A is 18.75% (3/16), experimental group B is 12.50%(2/16), experimental group C is 100%(16/16), there is significant difference between the groups (P<0.05); there is no significant difference between the experimental A, B group (P>0.05).Conclusion:HA/Ag composite coating is composed of Ag nanoparticles acicular HA and punctual, Ag uniform distribution of particles. The sustained release effect of composite materials can maintain effective antibacterial concentration and lower than the toxic concentration cell. Structure of the composite coating material with good compatibility. In vitro Against Staphylococcus aureus antibacterial rate of more than 95%. HA/Ag composite coating of artificial joint prosthesis animal body has good antibacterial activity Against Staphylococcus aureus.
