Objective:To study the genotyping of 5 strains of cryptococcus neoformans were isolated from cryptococcal meningitis and skin cryptococcosis in Xinjiang, and compared with the strains of mainland China.To determine the antifungal susceptibility in vitro to amphotericin B, itraconazole, fluconazole,5-fluorine cytosine, voriconazole, terbinafine, micafungin. Method:The genomic diversity were determined of five Xinjiag’s strains and 5 Shanghai’s strains isolated from cryptococcal meningitis were studied by polymorphic chain reaction random amplified polymorphic DNA method used random primers SEQ-6. Determining the strains in vitro antifungal susceptibility to amphotericin B, itraconazole, fluconazole,5-fluorine cytosine, voriconazole, terbinafine, micafungin used clinical and laboratory standards institute (CLSI) trace liquid dilution method (M27-A3).Results:Ten strains of cryptococcus neoformans divided into four genotypes, A, B, C, and D. One of Xinjiang strains showed type A, other 4 strains are type B.Five of Shanghai strains showed 3 kind of genotypes, B, C, and D. The 2 Xinjiang strains from meningitis and from skin cryptococcosis respectively showed same genotype B with a strains from Shanghai. The results showed that Xinjiang strains have highier homogeneous, and shanghai strains showed more genetic diversity. The MIC values of 5 Xinjiang’s strains to amphotericin B, itraconazole, fluconazole,5-fluorine cytosine, voriconazole, terbinafine, micafungin. are 0.5-2μg/ml、4-32μg/ml、32μg/ml、1-4μg/ml、0.5-1μg/ml、>16μg/ml respectively. All strains showed highest sensitivity to amphotericin B, medium sensitivity to azole type drugs, and low sensitive to terbinafine. Conclusions:Cryptococcus neoformans clinical isolates of the xinjiang region have unique genotype, and highier homogeneous. In vitro antifungal susceptibility test showed that Amphotericin B is best selections for cryptococcosis.
