[Objective] To confirm the existence of cryptococcus biofilm and explore the effect of cryptococcus biofilm on the mechanism of Cryptococcus drug resistance. [Method] We established both cryptococcus biofilm model in vitro and in vivo and observe its ultramicrostructure and metabolic activity. Analyze CSF and serum influence on cryptococcus biofilm. Susceptibility of Cryptococcus neoformans Biofilms to antifungal agents. [Result] The model of cryptococcus biofilm in vitro and in vivo showed cryptococcus biofilm was organic three-dimensional structure .Metabolic activity difference of cryptococcus biofilm in vitro at various time points during biofilm formation has statistical significance and kept stabilization until biofilm mature. Activity of biofilm formation were different for different serum type C.neoforman. Antifungal resistance of biofilm-grown cells increased in conjunction with biofilm formation. [Conclusion] The exsistence of C.neoforman biofilm were proved. C.neoforman biofilm can reduce the susceptibilities of antifungal agents.
