Objective: To study the synergistic effect of 5-Aza-cd R on bladder cancer T24 cells treating by mitomycinMethod: With the same concentrations of mitomycin(0.01mg/ml) and 5- ’aza-2 deoxycytidine(0, 0.25, 0.5, 1, 5, 10umol/L) treatment of bladder cancer cell line T24 48 hours, group 1 as control group(without mitomycin and 5- aza-2 deoxycytidine’). Cells in each group were used for the detection of Absorbance values, cell apoptosis was detected by flow cytometry, the expression of P-gp, MRP protein was detected by Western blot.Results: MTT detection of T24 cell proliferation, and single MMC group and combined with 5-Aza-cd R group, the combination of the two drugs on bladder cancer T24 cells and the A value of the differences were statistically significant(P < 0.01), the combination of the two drugs on bladder cancer T24 cells growth inhibition increased obviously, a synergistic effect. Cell apoptosis was detected by flow cytometry, cell apoptosis rate of each group are: 3.23 + 0.08%, 4.85 + 0.04%, 8.81 + 0.06%, 14.26 + 0.11%, 21.14 + 0.05%, 28.46 + 0.04%, 38.17 + 0.07%, apoptosis cells increased gradually, each group compared with the control group, the difference was statistically significant(P < 0.05). Detection of P-gp protein of Western Blot, the sample IOD value is: 451.71 + 0.08, 416.27 + 0.09, 315.42 + 0.17, 214.03 + 0.14, 174.71 + 0.23, 80.519 + 0.14, 38.322 + 0.19; detection of MRP group IOD value is: 438.73 + 0.05, 388.59 + 0.08, 322.53 + 0.13, 260.82 + 0.17, 227.53 + 0.04, 163.41 + 0.10, 128.23 + 0.21. Expression of P-gp, MRP protein of each group cells gradually decreased gradually increased, groups compared with the control group, the difference was statistically significant(P < 0.05).Conclusion: 5- Aza- cd R combination with mitomycin can inhibit T24 bladder cancer cell proliferation, promote cancer cell apoptosis and down-regulated the expression of tumor drug resistance related proteins(P- gp, MRP), and all of these were in a dose manner after treatment with increasing 5-Aza-cd R.
