Objective: Evaluat the influence of physical and neural activities on their own and their offspring rats after rats exposure to perfluorooctane sulfonate(PFOS) during pregnancy.Methods:120 healthy females with similar weight,40 male rats numbered from 1 to 120 and 1 to 40 by weight, respectively. From fifth rows and fifth columns to began,continuous take 120, 40 two digit,respectively.According to the random number and the order from small to large numbered these digit, if there appeared same number, first appears as a small.Set in advance: female rats numbered 1-30 and male rats numbered 1-10 corresponding to control group,female rats numbered 31-60 and male rats numbered 11-20 corresponding to low concentration group,female rats numbered 61-90 and male rats numbered 21-30 corresponding to middle concentration group,female rats numbered 91-120 and male rats numbered 31-40 corresponding to high concentration group.120 females, 40 male rats were divided into four groups randomly according to the above method.After being fed for 3 days, test parental rat neural activity level consistent whether or not and eliminate abnormal individuals through five days of water maze experiment.Between the rest parental rats in each groups, there were no differences between neural activity level(P < 0.05). days later, Male and female rat mated for 7 days, and then then kissed the male rats. From the first mated day to pregnancy day ended,with different doses of PFOS(control group: 2%-tween80 that is PFOS 0.0mg/(kg.d) low concentration group: PFOS 0.4mg/(kg· d), medium concentration group: PFOS 0.8mg/(kg · d), the high concentration group: PFOS 1.6mg/(kg · d)) by gavaged pregnant rats every day. And stop to gavage in Lactations(Postnatal day, PND),at the end of lactation(breast-feeding for 1 month),after iliac vein blood, killed all parent rats and 2-4 offspring rats(according to the actual sex case, try the best to left 1 male and 2 females each generation, and then filial generation rats were killed at 1 female and 1 male.these male and female rats’ weight differences between individuals within 10 g, respectively). Then collected the liver, thymus, testis, ovary, uterus, weighted these tissues, calculated these tissues’ organ coefficient, during feeding process, recorded the number of pregnant rats, the change of food intake, body weight and gestational age during pregnancy, parental intake, parental body weight changes in lactation time offspring rats’ weight in 1, 4, 7, 14, 21, 28 day in lactation time, After 1 month, recorded residual rats’ weight weekly.observe the he time of eyes open,the time of prick ear. recorded the birth number of offspring rats on PND1, during feeding process, recorded offspring rats’ survival,mortality,deformity in 1, 4, 7, 14, 28 day.After 1 month, recorded the number of survival,death and malformation of the remaining rats.In the generation of 2 months,According to the same method of parental rats, by five days of water maze experiment,testing son generation rats nerve activity level is difference wither or not on the basis of Parental rats,neural activity level consistent. 7 days after the water maze, close cage son generation 7 days,no longer infected son generation during pregnancy,Record the gestational age time, the number of pregnancy, the litter size of son generation.Record the first day,s survival and deaths deformity number.Results:①The influence of the parental rats:From the 6 days to 21 days of pregnancy,each time point,parental rats, average body weight of P values were 0.029,0.03,0.004,0.008,0.003,0.004 respectively,P values < 0.05.At 18 days of food intake,the control group and the high concentration of the average intake,s P value is 0.022,;low concentration group compared with concentration in the group and high concentration groups, P values were 0.047, 0.004.1 months of parental lactation intake,P values<0.001.Pregnancy length P=0.013.②The influence of pair generation rats:the birth rate of high concentration group was 17.69%( ?2 was 36.606,P=0.000),lower than the other 3 groups on PND1. The death of the control group were 11、5、95,the death rate of the control group were 4.78%,2.28%,41.30% on PND1, PND4 and in PND1-14 respectively,( ?2 were 16.106、37.459、9.473 respectively,P,were <0.025)lower than the low concentration group and high concentration group. This study has found no monster and postnatal malformation.There were no differences in mean body weight of each group in the first 7 days, but after 7 days, the average body weight of control group were lower than the 3 experimental groups.the time of eyes open and prick ear, control group’s were mainly in the postnatal fourteenth days and sixteenth days, earlier compared with3 experimental groups(P<0.025).There was no difference in female rats’ organ coefficient, control group’s organ coefficient of testis in male was lower than concentration and high concentration group’s(P<0.05). And there were no difference with other organ coefficient.③ the influence of the second generation:The first day,s mortality of the second generation, control group compared with three group,P<0.001.The fifth day,three experimental mortality was 100%, while the control group was only 10%.Conclusion:In this experiment, after rats exposed to PFOS in pregnancy, there caused a certain influence in their body weight, food intake, organs of rats.At the same time because of PFOS can through the placental barrier and PFOS have chemicals, which lead to Son generation of rats has a very big influence in each physiology and reproduction,and the rat survival generation has received the serious threat,And because of PFOS,s chemicals, as the steady accumulation of PFOS after infected during pregnancy,Its influence over time become more and more obvious.Especially its impact on children should not be ignored,So we should try to reduce PFOS exposure during pregnancy.
