Effect Of Sini Decoction On AngⅡ,TGF-β1 And CTGF Of Myocardial Fibrosis Rats With Abdominal Aorta Banding

Objective To study the intervention of Sini decoction on myocardial fibrosis rats with abdominal aorta banding, and explore its possible mechanisms of the intervention.Methods Forty-eight 8 weeks old male SD rats were randomly divided into sham operation group(A), model group(B), captopril group(C) and Sini decoction group(D), 12 rats in each group. The models were established by partially banding of abdominal aorta according to Doering’s method. The fourth day after operation, group A and group B were were orally administered with distilled water, group C was perfused with captopril(100 mg/kg), group D was administered with Sini decoction(3.8 g/kg), all rats were prefused once in a day, and lasted for 8 weeks. 8 weeks later, heart weight indexes were calculated, hemodynamics changes of heart were tested, the changes of myocardial tissue morphology were observed by hematoxylin-eosin(HE) staining and Masson staining, and myocardial collagen volume fraction(CVF) was calculated, ELISA kit was used to measure the concentration of angiotensin Ⅱ(AngⅡ) in serum, the expression of transforming growth factor-β1(TGF-β1) and connective tissue growth factor(CTGF) were tested by Immunohistochemistry method and Western blotting method.Results Compared with group A, heart weight index, collagen volume fraction of myocardium, AngⅡ levels in serum, and the expression of myocardial TGF-β1 and CTGF in group B significantly increased(P<0.05). Compared with group B,heart weight index, collagen volume fraction of myocardium, AngⅡ levels in serum, and the expression of myocardial TGF-β1 and CTGF significantly reduced in group C and D(P<0.05), but much higher than group A. There was no statistic significance between group C and D(P>0.05).Conclusions Sini decoction can reduce Ang levels and inhibit the expression ofⅡ TGF-β1 and CTGF in myocardium, which may explain its mechanism of amelioration effect on myocardial fibrosis.
